摘要
目的建立三桠苦中山柰酚的HPLC含量测定方法。方法采用反相高效液相色谱法。色谱柱:Diamonsil C18柱(250 mm×4.6 mm,5μm);流动相:甲醇-0.4%H3PO4(体积比55∶45);流速:1 mL.min-1;检测波长:368 nm;柱温:25℃;进样量:10μL。结果山柰酚在0.48~4.80μg范围内与峰面积呈良好线性关系(r=0.999 4),平均回收率为98.52%,RSD为2.45%(n=6)。结论该方法简便、准确、分离度好,可用于三桠苦中山柰酚的含量测定。
Objective To establish a HPLC method for the determination of kaempferol in Melicope ptelefolia(Champ.ex Benth.) Hartley.Methods RP-HPLC method on C18 column(250 mm×4.6 mm,5 μm) was used with the mobile phase consisted of methanol and 0.4% phosphoric acid.The flow rate was 1.0 mL·min-1.The UV detection wavelength was 368 nm.The column temperature was maintained at 25 ℃ and the injection volume was 10 μL.Results The linearity of kaempferol was good in the range of 0.48-4.80 μg(r=0.999 4),and its average recovery was 98.52%,RSD=2.45%(n=6).Conclusion This method is simple,accurate,reliable with good resolution and can be used for the quantitative determination of kaempferol in M.ptelefolia.
