摘要
研究了聚酰胺树脂对血竭中龙血素A和龙血素B的提取效果。采用HPLC进行定量分析,考察了上样方法、洗脱剂体积分数、洗脱流速、树脂使用次数等因素对提取效果的影响,并确定了适宜的梯度洗脱方式。结果表明,将血竭样品用乙酸乙酯溶解后,与聚酰胺树脂混合,然后将溶剂蒸发完全进行吸附,树脂的吸附量为1g.(g树脂)-1。在0.8mL.min-1洗脱流速下,7BV的50%乙醇可梯度洗脱被聚酰胺树脂吸附的龙血素A和龙血素B,解吸率均达91%以上,聚酰胺树脂使用3次后即需再生。该工艺收率高、操作简便,适于血竭龙血素的提取。
The abstraction of loureirin A and B in dragon's blood by polyamide resin was studied. The effect of sampling method, eluent volume fraction, eluting velocity, frequency of use of polyamide resin on the abstrac- tion efficiency were investigated,and the quantitative analysis was conducted on HPLC. And the gradient elu- tion way was optimized. Results showed that,the adsorbing capacity of polyamide was 1 g · g-1 resin when the process was as follows.after dissolved with acetic ether, the sample was mixed with polyamide resin,and then the organic solvent was evaporated completely. The loureirin A and B adsorbed by polyamide resin could be gra- diently eluted more than 91% by 7 BV of 50% ethanol at the eluting velocity of 0.8 mL·min-1. The process was fit for the abstraction of loureirin A and loureirin B in dragon's blood with high yield and simple operation.
