人脐带间充质干细胞移植对四氯化碳致肝硬化大鼠ALB的影响

摘要 目的探讨人脐带间充质干细胞移植至四氯化碳所致的肝硬化大鼠体内后,肝组织和血清中白蛋白的表达情况。方法随机选取健康雄性SD大鼠8只作为正常组,另同样方法选取35只SD大鼠给予40%CCl4溶液每周2次腹部皮下注射,用来制备肝硬化大鼠模型,造模成功后,随机分为肝硬化组和HU-MSCs移植组,HU-MSCs移植组给予HU-MSCs1×107经尾静脉移植,移植后3周处死所有大鼠,进行免疫组织化学检测抗人表皮生长因子受体(EGF-R)的表达,免疫荧光和Western blo法以及RT-PCR法检测肝组织中ALB的表达,并于生化检验室检测血清中ALB的含量。结果正常组和肝硬化组中均未发现阳性表达的抗人EGF-R存在,HU-MSCs移植组中发现较多抗人EGF-R的表达;肝组织中ALB的表达较肝硬化组明显增加,与正常组无明显差异。结论 HU-MSCs移植到肝硬化大鼠体内,能够长期存活,并改善ALB的表达和分泌。 【Objective】To explore the human umbilical cord mesenchymal stem cells transplantation into mice liver cirrhosis caused by carbon tetrachloride,and test serum albumin expression in serum and liver tissue.【Methods】Select 8 health male SD rats randomLy as normal group and the others as the same methods select 35 SD rats to give 40% CCl4 solution 2 times per week in the subcutaneous injection,to make liver cirrhosis rats model,after the success of the model,randomLy divided into liver cirrhosis group and HU-MSCs transplantation group,HU-MSCs transplantation group give HU-MSCs 1 x 107 from the tail vein,three weeks after transplantation to suicide all the rats,immunohistochemical test against human epidermal growth factor receptor(EGF-R) expression,the Western blot test immunofluorescence and in the expression of liver tissue ALB,and in the biochemistry checkout room serum ALB detection of content.【Resluts】The normal group and liver cirrhosis group were not found the expression of positive EGF-R exist,HU-MSCs transplantation group found many positive cells expression EGF-R;In the expression of liver tissue ALB liver cirrhosis group is significantly increased,and compared with the normal group,that is no obvious difference.【Conclusion】HU-MSCs transplantation into the liver cirrhosis mice,can live for a long time,and improve the expression of ALB and secretion.
