固本止咳中药对慢性阻塞性肺疾病小鼠肺功能及呼吸道分泌sIgA的影响 被引量:20

Effect of Gubenzhike recipe on pulmonary function and sIgA in respiratory tract of COPD mice

摘要 目的:观察固本止咳中药对慢性阻塞性肺疾病(COPD)模型小鼠肺功能及呼吸道分泌分泌型免疫球蛋白(sIgA)的影响。探讨固本止咳中药治疗COPD的作用机制。方法:采用鼻腔滴入脂多糖(LPS)加熏香烟的方法建立COPD小鼠模型,实验分为空白对照组、模型对照组、固本止咳中药组;运用HE染色、电镜对形态学进行观察,评价COPD小鼠模型;使用动物肺功能仪分析系统检测小鼠肺功能;应用ELISA方法检测小鼠气道sIgA分泌水平。结果:COPD小鼠模型形态学结果符合COPD临床病理形态学改变;肺功能实验显示FEV0.1、FEV0.2、FEF25-75、PEF模型对照组较空白对照组显著性降低(P<0.05),固本止咳中药组较模型对照组显著性增高(P<0.05);气道sIgA分泌水平模型组较空白对照组显著性降低(P<0.05),固本止咳中药组较模型对照组显著性升高(P<0.05)。结论:固本止咳中药对COPD模型小鼠小气道功能及局部黏膜状态有一定的改善作用。 Objective: To observe the effect and mechanism of Gubenzhike recipe on pulmonary function and sIgA level in respiratory tract of COPD mice.Methods: The mouse COPD model was established by exposure to cigarette smoke in combination with intra-nostrils injection of lipopolysaccharide.The mice were divided into three groups: negative control group,model group and Gubenzhike recipe group;Use the methods of HE eosin staining and electronic microscope to observe the morphology characteristic of COPD model;Test the pulmonary function by animal pulmonary function machine;Use the ELISA to test the sIgA level in respiratory tract.Results: The morphology characteristic is up to COPD;The FEV0.1,FEV0.2,FEF25-75,PEF and the content of sIgA in the upper respiratory tract in Gubenzhike recipe group had significant difference compared with those in model group(P0.05).The negative control group had no significant difference compared with Gubenzhike recipe group.Conclusion: Gubenzhike recipe might improve the pulmonary function and respiratory tract mucosa immunity of COPD model.
