摘要
目的探讨水通道蛋白4(AQP4)在大鼠脑缺血再灌注后的表达及丹参川芎嗪的干预作用。方法选择健康大鼠100只,随机分为假手术组,模型组,低剂量药物干预组(低剂量组),高剂量药物干预组(高剂量组),每组25只。各组大鼠又按术后时间(6、24 h和3、5、7 d)分为5个亚组,每亚组5只。分别对大鼠进行Longa评分,干湿重法测定脑组织含水量;免疫组织化学法检测脑组织AQP4的表达。结果术后大鼠行为异常,3 d最明显,7 d恢复至术前水平。在同时间点,与假手术组比较,低剂量组、高剂量组和模型组大鼠脑组织含水量、AQP4表达明显升高(P<0.01);与模型组比较,低剂量组、高剂量组脑含水量、AQP4表达明显降低(P<0 01),高剂量组明显低于低剂量组(P<0.01)。而模型组、低剂量组、高剂量组各时间点比较,差异有统计学意义(P<0.01)。AQP4表达水平与脑含水量呈正相关(,=0.871,P<0.01)。结论脑缺血再灌注后,脑组织AQP4的表达变化与脑水肿的动态变化存在时间上的一致性;丹参川芎嗪可以改善大鼠神经缺损症状,减轻脑水肿,降低AQP4表达水平。
Objective To study the expression of AQP-4 in rats following cerebral I/R and its intervention with Radix Salviae Miltiorrzhizae and Ligustrazin. Methods One hundred healthy SD rats were randomly divided into sham operation group, model group, low-dose drug treatment group and high-dose drug treatment group(n= 25). The rats in each group were further divided into 5 subgroups according to different time points(6 h,24 h,3 d,5 d,7 d)after operation(n=5). The rats were scored according to the Longa scale. Water content in brain was measured using the wet-dry method. Expression of AQP-4 in brain tissue was detected with immunohistochemistry staining. Results The behaviors of rats were abnormal, especially on day 3 after operation,and became normal on day 7 after operation. The water content and AQP-4 expression level in brain tissue were significantly higher in lowand hogh-dose drug treatment groups and model group than in sham operation(P〈0.01)and significantly lower in lowand hogh-dose groups than in model group, and in high-dose drug treatment group than in low-dose drug treatment group at the same time point(P〈0.01). The AQP-4 expression level was positively related with the water content in brain (r=0. 871,P〈0.01). Comclusion AQP-4 expression in brain tissue is related with cerebral edema following cerebral I/R. Radix Salviae Miltiorrzhizae and Ligustrazin can improve the nerve defect symptoms and cerebral edema,and reduce the AQP-4 expression level.
