摘要
目的通过研究缩泉丸对肾虚多尿大鼠肾脏AT1R mRNA及蛋白表达的影响,阐明其补肾缩尿的分子药理学机制。方法采用腺嘌呤造成大鼠肾虚多尿模型,利用酶联免疫分析法检测各组动物血中AngⅡ含量;并采用RT-PCR、免疫组化的方法检测各组动物肾脏AT1R mRNA及蛋白的表达。结果与模型对照组比较,缩泉丸可显著提高肾虚多尿大鼠血中AngⅡ含量,并可上调模型大鼠肾脏AT1Rm RNA及蛋白的表达。结论缩泉丸可通过促进肾脏AT1RmRNA及蛋白的表达,增强AngⅡ的功能,发挥调节水液代谢的作用。
Objective To expound its mechanism of nourishing kidney and reducing urine by discussing the influence of SuoQuan capsule on the mRNA and protein expressions of AT1R. Methods Employing adenine to build the model of deficiency of the kidney and diuresis rats, inspecting the contents of Ang ]I in blood , mRNA and protein expressions of AT1R with ELISA , RT- PCR and IHC technology among model group, SuoQuan capsule group, JinSuoGuJinWan group and control group. Results Compared with model group , SuoQuan capsule could remarkedly increase the contents of Ang ]I in blood and the expressions of mRNA and protein of model rats AT1R. Conclusion SuoQuan capsule can adjust water metabolize of body by increasing the expressions of mRNA and protein of AT1R.
