摘要
目的:研究妇科再造丸(FKW)对子宫肌瘤模型大鼠的保护作用。方法:实验分为6组,即正常对照、模型、桂枝茯苓胶囊(0.3g·kg-1)与FKW高、中、低剂量(1.12、0.56、0.28g·kg-1)组。雌二醇(E2)、孕激素(P)负荷法复制大鼠子宫平滑肌瘤样增生模型,观察子宫组织病理改变,检测子宫组织中雌激素受体(ER)、孕激素受体(PR)表达与血清中E2、P水平。结果:与正常对照组比较,模型大鼠较正常大鼠子宫明显增大,血清中E2、P水平显著升高,子宫组织中ER、PR的表达显著增强(P<0.01);与模型组比较,FKW高、中、低剂量组血清中E2、P水平显著降低,FKW高剂量组平滑肌细胞ER、PR的表达显著减弱(P<0.05或P<0.01)。结论:FKW有抗大鼠子宫平滑肌瘤样增生的作用,其机制可能与调节血中E2、P水平,抑制子宫平滑肌细胞ER、PR的表达有关。
OBJECTIVE: To study the protective effects of Fuke zaizao pills (FKW) on hysteromyoma model rats. METHODS: Model rats were divided into 6 groups, i.e. normal control group, model group,Guizhi fuling capsule group (0.3 g·kg-1) and FKW high-dose, medium-dose and low-dose groups(1.12,0.56,0.28 g·kg-1). The uterine leiomyoma-like hyperplasia model rats were induced by estrogen and progesterone load method. The pathological changes of uterus were observed. The expression of estrogen and progesterone receptors in smooth muscle cells and the level of serum estrogen and progesterone were measured. RESULTS: Compared with normal control group, hysterauxesis of model group was more significant, and the serum level of E2 and P and the expression of ER and PR in the model group increased obviously(P0.01). Compared with model group, the serum level of E2 and P in FKW high-dose, medium-dose and low-dose groups decreased significantly, and the expression of ER and PR of smooth muscle cells decreased significantly (P0.05 or P0.01). CONCLUSION: The results suggest that FKW have the function of resisting uterine leiomyoma-like hyperplasia in rats, which may be related to the regulation of serum level of E2 and P,the inhibition of the expression of ER and PR of smooth muscle cells.
