摘要 目的观察重组人半乳糖凝集素-3(Galectin-3)对干细胞源性Tip血管内皮细胞增殖能力的影响。方法密度梯度离心法提取人外周血内皮祖细胞,并将其定向诱导为Tip血管内皮细胞。培养液中分别加入不同终质量浓度的Galectin-3(0、0.1、1.0、2.5、5.0、10.0mg/L)培养24h。噻唑蓝(MTT)比色法观察不同浓度Galectin-3对外周血内皮祖细胞源性Tip血管内皮细胞增殖能力的影响。结果6组不同浓度Galectin-3作用于Tip血管内皮细胞后的吸光度(A)值分别为(0.103±0.068、0.122±0.027、0.139±0.023、0.289±0.042、0.912±0.146、1.188±0.337)。Galeetin-3为5.0mg/L及10.0mg/L浓度组的A值分别为明显高于0、0.1、1.0及2.5mg/L浓度组,差异有统计学意义(P〈0.05);10.0mg/L浓度组A值明显高于5.0mg/L浓度组,差异有统计学意义(P〈0.05)。结论Galectin-3可促进体外培养干细胞源性Tip血管内皮细胞的增殖能力。 Objective To'observe-the effects of Galectin-3 on proliferation of endothelial Tip cells derived from stem cells. Methods The cultured peripheral blood endothelia!progenit0r cells in vitro were isolated and purified from human peripheral blood, and the cells were induced to differentiate into endothe- lial Tip cells. Then, the cells were cultured with Galectin-3 (0, 0. 1, 1.0, 2. 5, 5.0, 10. 0 mg/L) for 24 h. The proliferation of endothelial Tip ceils derived from peripheral blood endothelial progenitor cells was measured by using methyl thiazol tetrazolium (M3T) assay. Results The absorbance (A) values in Ga- lectin-3 groups (0.0, 0.1, 1.0, 2.5, 5.0, 10.0 mg/L) were (0.103_+0.068), (0.122 +0.027), (0. 139 -+0. 023), (0. 289 +0. 042), (0. 912 +_0. 146), and (1. 188 +_0. 337) respectively. The A val- ues in Galectin-3 (5.0 and 10. 0 mg/L) groups were significantly higher than in galectin-3 (0, 0. 1, 1.0 and 2. 5 mg/L) groups, P 〈 0. 05. There was significant difference in the A values between 5.0 mg/L group and 10. 0 mg/L group,P 〈 0. 05. Conclusion Galectin-3 can promote the proliferation of endothelial Tip ceils derived from stem cells.
