大肠癌中转录因子激活蛋白-2α与癌胚抗原的表达及其相关性

Expression of transcription factor AP-2α and CEA in colon cancer and the correlation between the two factors

摘要 目的探讨转录因子激活蛋白-2α(AP-2α)与癌胚抗原(CEA)在大肠癌发生、发展过程中的表达情况及其表达的相关性。方法采用实时荧光定量PCR的方法分析60例大肠癌组织及正常组织中AP-2α mRNA及CEAmRNA表达差异,分析其与患者临床资料、病理参数之间的关系,研究60例大肠癌组织中AP-2αmRNA及CEAmRNA表达的相关性。结果大肠癌组织、正常组织AP-2α mRNA相对表达值(RQ)分别为3.264±0.877、17.484±3.608;CEAmRNARQ值分别为16.421±4.987、2.219±1.313;大肠癌组织中AP-2α mRNA的表达显著低于正常组织,CEAmRNA显著高于正常组织,差异均有统计学意义(P〈0.01);大肠癌组织AP-2α mRNA和CEAmRNA表达情况在不同性别、年龄、肿瘤部位中差异均无统计学意义(P〉0.05);大肠癌的不同组织学分级及Dukes分期中。CEA呈现出过表达状态,而AP-2α呈现出表达降低或缺失状态;60例大肠癌组织中AP-2α和CEA的表达具有相关性(r=-0.790,P〈0.01),且呈负相关关系(-1〈r〈0)。结论转录因子AP-2α在大肠癌组织中呈现低表达或表达缺失状态,AP-2α和CEA的表达呈现一定的负相关关系,提示AP-2α和CEA可为肿瘤的诊断和治疗提供新的依据。 Objective To investigate the expression of transcription factor AP-2c~ and CEA in the process of occurrence and development of colorectal cancer and the correlation between the two tran- scription factors. Methods The expression levels of AP-2α and CEA mRNA in 60 colon cancer tissues and corresponding normal tissues were detected by Real-time PCR and the results with the clinical features and pathological characters were analyzed. The relationship between the expression of AP-2α mRNA and CEA mRNA in 60 colon cancer tissues was analyzed. Results The expression levels of CEA mRNA was higher in colon cancer tissues than that in the matched normal tissues (P 〈 0. 01 ) , whereas the expression levels of AP-2α mRNA in the colon cancer tissues was significantly lower than that in the matched normal tissues (P 〈 0.01 ). There was no significant correlation between AP-2α, CEA mRNA expression levels and age, sex, tumor location (P 〉0. 05). Loss expression or down regulation expres- sion of AP-2α mRNA was detected, whereas CEA mRNA was highly expressed in the different histologi- cal grade and Dukes stages. The expression of AP-2α mRNA and CEA mRNA was positively correlated with the histological grade in colon cancer. A negative corre AP-2α mRNAandCEAmRNA(r = -0. 790, P 〈 0. 0l on was found between the expression of Conclusions The expression of AP-2α mRNA is down-regulated or lossed, whereas CEA is highly expressed in colon cancer. There is negative correlation between the expression of AP-2α mRNA and CEA mRNA, indicating that AP-2α and CEA may provide the new clues of diagnosis and treatment of tumor.
