摘要
目的考察不同制酸类中药对胃黏膜氨基己糖及前列腺素(PGE2)的影响。方法选择雄性Wistar大鼠,随机分为正常组、模型组,以及瓦楞子、白螺丝壳、牡蛎、海螵蛸、浙贝母低、中、高剂量组。各药物组予灌胃相应药物,正常组、模型组灌胃等容积蒸馏水;于药物干预6 d后一次性灌服75%乙醇制备大鼠急性胃溃疡模型,取样测定胃黏膜溃疡指数及氨基己糖、PGE2含量。结果 5种临床常用的制酸类中药均可明显降低乙醇所致大鼠急性胃溃疡模型的溃疡指数。牡蛎中剂量组、海螵蛸中剂量组和浙贝母高剂量组氨基己糖含量较模型组明显升高(P<0.05);而白螺丝壳中、高剂量组和海螵蛸高剂量组的氨基己糖含量较模型组更高,差异有统计学意义(P<0.01)。白螺丝壳中剂量组,牡蛎低、高剂量组PGE2含量较模型组明显升高(P<0.05);而瓦楞子中剂量组,牡蛎中剂量组,海螵蛸低、中剂量组PGE2含量较模型组升高,差异有统计学意义(P<0.01)。结论 5种制酸类中药均可降低75%乙醇所致大鼠急性胃溃疡模型的溃疡指数,对乙醇所致胃黏膜损伤有明显的保护作用;在胃黏膜保护机制方面,5种制酸类中药的不同剂量对氨基己糖及PGE2水平的作用存在一定的差异。
Objective To investigate the effects of gastric acid-inhibiting medicines on hexosamine and PGE2 in gastric mucosa.Methods The gastric mucosal lesion in male Wistar rats induced by 75% ethanol;the index of ulcer,the concentration of hexosamine and PGE2 in gastric mucosa of different dosages of the five different Chinese medicines for inhibiting gastric acid.Results The five different Chinese medicines for inhibiting gastric acid(including Cuttlefish bone,Concha Arcae,Concha Ostreae,Spiral shell and Bulbus Fritillaria Thunbergii) had preventive and therapeutic effects on gastric mucosal lesion induced by 75% ethanol;the secretion of hexosamine in Concha Ostreae(the middle-dose) group,Cuttlefish bone(the middle-dose) group and Bulbus Fritillaria Thunbergii(the high-dose) group were enhanced significantly as compared with that of the model group(P0.05);and the secretion of hexosamine in Spiral shell(the middle-,high-dose) groups and Cuttlefish bone(the high-dose) group were enhanced significantly as compared with that of the model group(P0.01);the concentration of PGE2 in Concha Arcae(the middle-dose) group,Cuttlefish bone(the low-,middle-dose) groups increased significantly as compared with that of the model group(P0.01);the concentration of PGE2 in Spiral shell(the middle-dose) group,Concha Arcae(the low-and high-dose) groups increased significantly as compared with that of the model group(P0.05).Conclusion The five different Chinese medicines for inhibiting gastric acid have preventive and therapeutic effects on gastric mucosal lesion induced by ethanol,and different doses of different drugs prevent gastric mucosa in different mechanisms.
