摘要
研究软枣猕猴桃粗蛋白酶在不同条件下分解肌动球蛋白效果,通过SDS-聚丙烯酰胺凝胶电泳得出软枣猕猴桃粗蛋白酶分解肌动球蛋白的效果和反应最佳条件。结果表明:软枣猕猴桃粗蛋白酶浓度为0.28mg/mL,比活力为497.11U/mg,肌动球蛋白浓度为1.54mg/mL,软枣猕猴桃粗蛋白酶对肌动球蛋白具有强烈的分解作用,而且反应最佳条件是pH值为7.0、反应时间为2.5h、温度为60℃。
In this experiment, we studied the disintegration effects of crude proteases from Actinidia arguta on actomyosin under different reaction conditions and confirmed the best condition for crude protease disintegrating actomyosin with SDS-PAGE. Results showed that the concentration of the crude protease extracted from Actinidia arguta was 0.28 mg/mL, the enzyme activity was 497.11 U/mg, and the concentration of the actomyosin was 1.54 mg/mL. The crude protease extracted from Actinidia arguta had a strong disintegration effect on actomyosin and the optimum reaction condition included a pH of 7.0, a reaction time of 2.5 h, and a reaction temperature of 60 % respectively.
