芡实提取物对衰老小鼠脑组织p53蛋白表达的影响及机制 被引量:1

摘要 目的观察芡实提取物对D-半乳糖(D-gal)致衰老小鼠氧化系统指标及p53蛋白表达的影响,探讨芡实提取物延缓衰老的机制。方法 104只昆明雄性小鼠随机分为正常对照组,D-gal模型组,芡实乙醇提取物高、低剂量组,芡实乙酸乙酯提取物高、低剂量组,芡实正丁醇提取物高、低剂量组。各组用相应药物连续处理6 w。用比色法检测小鼠脑组织总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)的活性。用HE染色法和免疫组织化学染色法检测小鼠脑组织海马区细胞形态和p53蛋白表达水平。结果芡实提取物能有效提高衰老小鼠脑组织T-AOC、SOD和GSH-Px水平(P<0.05,P<0.01),降低MDA含量(P<0.05,P<0.01),且呈量效依赖关系,还能改善海马区神经细胞受损状态,下调p53蛋白的表达(P<0.05)。结论芡实提取物可下调p53蛋白的表达,其机制可能与清除自由基及改善脑组织海马区神经细胞受损情况有关。 Objective To investigate the deferring aging effect and the associated mechanisms of Euryale ferox seed extracts on indexes of oxidation system and the expression of p53 protein in the brain of aging mice induced by D-gal. Methods 104 male Kunming mice were divided randomly into the normal control, D-gal model, high and low dose of ethanol, acetic ester and n-butanol extracts from Euryale ferox seed groups. All animals were administered by corresponding medicine for 6 weeks. The activities of total antioxidant capacity (T- AOC), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) in brain were detecte& HE stain and immunohistochemistry techniques were performed to determine the form of hippocampal nerve cells and the expression of p53 protein in the brain choroid. Results Compared with the model group, Euryale ferox seed extracts could increase the activities of T-AOC, SOD and GSH-Px (P 〈 0.05, P 〈 0. 01 ), reduce MDA ( P 〈 0. 05, P 〈 0. 01 ) with dose-dependent tendency, prevent hippocampal nerve cells from damaging obviously and down-regulate the expression of p53 protein (P 〈 0. 05). Conclusions Euryale ferox seed extracts can down-regulate the expression of p53 protein. The mechanisms may be related to scavenging free radicals and preventing hippocampal nerve cells from damaging.
