2型糖尿病合并冠心病患者载脂蛋白E基因多态性研究 被引量:6

摘要 目的 :研究载脂蛋白 E(Apo E)基因多态性与 2型糖尿病患者合并冠心病的关系。方法 :用聚合酶链反应 -限制性片段长度多态性 (PCR- RFL P) ,研究了 12 5例 2型糖尿病 (DM)患者和 71例健康体检者 Apo E基因型 ,分析了 Apo E基因多态性与 2型糖尿病及其合并冠心病的关系。结果 :(1)糖尿病组 (DM)、糖尿病合并冠心病组 (DC)及对照组等位基因频率分别为 5 .5 %、89.7%、4.8% ,4.1%、85 .3%、10 .6 %和 5 .6 %、86 .6 %、7.8%。相互间 P均 >0 .0 5 ,说明 Apo E基因型与中国人的 2型糖尿病及合并冠心病的发生发展无相关性。 (2 )通过对不同 Apo E基因型的血胆固醇 (TC)、甘油三酯 (TG)、高密度脂蛋白 (HDL )及低密度脂蛋白 (L DL )水平比较 ,发现携ε4等位基因的个体血 TC高于携其他 ApoE等位基因的个体 ,而携ε2等位基因的个体血 TG的水平高于携ε3、ε4等位基因的个体 (P<0 .0 5 )。 (3)Apo E基因多态性与高血压 (HT)无显著相关性。 (4 )携ε2、ε3、ε4等位基因的个体之间冠心病的发生率差异无显著性。结论 :Apo E等位基因多态性不是 Objective:To Study the relationship between the polymorphism of apolipoprotein E(Apo E)gene and type 2 diabetes mellitus and its carkiovascular complications.Methods:Apo E genetype was determined by polymerase chain reaction restriction fragment length polymorphism(PCR RFLP) in 125 type diabetes mellitus inpatients and 71 normal controls.Results:The frequencies of;ε2,ε3,ε4 alleles in these three groups were 5.5%,89.7%,4.8%;4.1%,85.3%,10.6% and 5.6%, 86.6%, 7.8%( P >0.05).It was suggested that no correlation between Apo E genotypes and type2 diabetes mellitus in Chinese had been found.Comparing the levels of serum lipid among type 2 diabetes mellitus with different Apo E genotype,patients carrying ε4 allel were associated with increased concentrations of total cholesterol,it was the same between the patients carrying ε2 allele and the level of triglyceride.The incidence of coronary heart disease or hypertension in type 2 diabetes mellitus patients had no relationship to Apo E gene polymorphism.Conclusions:these findings suggest that Apo E gene polymorphism is not a risk factor of coronary heart disease in type 2 diabetes mellitus. [
