胃癌miRNAs表达谱及胃癌组织miR-21表达临床意义探讨 被引量:7

miRNAs expression profiles and the clinical significance of miR-21 expression in human gastric cancer tissues

摘要 目的:筛选胃癌miRNAs表达谱,探讨miR-21在胃癌组织中的表达情况及其临床病理意义。方法:收集手术切除新鲜胃癌标本共65例,包括胃癌组织及相应胃正常上皮组织,随机选取其中6例,应用miRNA芯片技术研究胃癌的miRNAs表达谱。采用实时定量PCR验证芯片结果并检测miR-21在65例胃癌组织标本及相应胃正常上皮组织的表达水平,探讨其表达与胃癌临床病理参数间的关系。结果:与胃正常上皮组织相比,胃癌组织中表达上调超过2倍的miRNAs有7个(miR-374b*、miRPlus-E1212、miR-338-5p、miR-297、miR-21、miR-135b和miR-18a),表达下调超过2倍的miRNAs有9个(miR-29b-2*、miR-1260、miRPlus-E1241、miR-S1-5p、miR-148a、miR-29c、miR-647、miR-196b*和ebv-miR-BART5)。46例(70.8%)胃癌组织中miR-21表达水平明显高于胃正常上皮组织,且与肿瘤的分化程度(χ2=6.257,P=0.012)、浸润深度(χ2=5.705,P=0.017)和有无淋巴结转移(χ2=7.107,P=0.008)有关。结论:筛选出的差异表达miRNAs可能参与胃癌的发病;miR-21与胃癌生物学行为密切相关,可能成为胃癌诊断和治疗的潜在生物学指标。 OBJECTIVE:To study miRNAs expression profile and explore miR-21 expression and its clinicopathologi- cal significance in human gastric cancer tissues. METHOD: Sixty-five pairs of fresh gastric cancer tissues and correspond- ing normal gastric epithelium were collected. Six of them were randomly selected and miRNA microarray were applied. Quantitative real-time PCR were used to validate the reliability of chips and detect the expression of miR-21 in sixty-five cases of gastric cancer tissues and corresponding normal gastric tissues. RESULTS:Compared with normal gastric tissues, 7 miRNAs (miR-374b * , miRPlus-E1212, miR-338-5p, miR-297, miR-21, miR-135b and miR-18a) were significantly up- regulated more than 2 folds, 9 miRNAs (miR-29b-2 * , miR-1260, miRPlus-E1241, rniR-S1-5p, miR-148a, miR-29c, miR- 647,miR-196b * ,ebv-miR-BARTS) were significantly downreguated more than 2 folds in gastric cancer tissues. The re- sults of microarray were verified by quantitative real-time PCR. MiR-21 expression was significantly higher in most of gastric cancer tissues compared with corresponding epithelial tissues(46/65,70.8%). Its high expression was related to tumor differentiation (X^2= =6. 257,P=0. 012) ,invasion (X^2==5. 705,P=0. 017) and lymphnode metastasis (X^2= =7. 107, P=0. 008). CONCLUSIONS: Differential expressed miRNAs may be involved in gastric cancer and provides a new basis for research about development mechanism of gastric cancer;miR-21 is closely related to biological behavior of human gas- tric cancer,it could be an indicator in diagnosis and treament of human gastric cancer.
