摘要
目的 从薏苡仁中分离提取薏苡仁多糖 (Coixan) ,并观察其对多种模型小鼠血糖水平的影响。方法 ①采用热水提取、低温减压浓缩、三氯醋酸法去蛋白、流水透析、SephadexG 75柱层析等方法。②采用四氧嘧啶、肾上腺素尾静脉注射建立高血糖小鼠模型 ,尾静脉取血 ,血糖仪测定血糖水平。结果 ①被提取的薏苡仁多糖是鼠李糖、阿拉伯糖、甘露糖、半乳糖和葡萄糖组成 ,分子量约为 1.5× 10 4 ,产率为 0 .12 5 % ;②口服给予Coixan对正常小鼠无明显降血糖作用 ;腹腔给予Coxian剂量在 5 0mg/kg和 10 0mg/kg时 ,能降低正常小鼠、四氧嘧啶糖尿病模型小鼠和肾上腺素高血糖小鼠的血糖水平 (P <0 .0 5&P <0 .0 1)。且呈现一定的量效关系。结论 薏苡仁多糖对正常及高血糖模型小鼠均有降血糖作用 。
Objective To study the method of extraction of coixan, a polysaccharide from the seeds of coix lacchryma jobi var, ma yuen and its hypoglycemic action in mice. Methods ①The dried coix seeds were extracted with hot water and the extract was concentrated under lower pressure to get the crude coixan. Its protein content was removed with trichloracetic acid (TCA). Then the crude coixan was dialyzed with cellulose tubings against flowing water for 7 d and chromatographed over sephadex G 75 with a column of (0-1)mol/L NaCl to obtain purified coixan. ②The model of diabetes mellitus was established with alloxan injection through the tail vein in mice and the mode of hyperglycemia with adrenaline injection in mice too. Blood samples were collected with tail vein puncture to determine blood glucose level. Results ①Refined coixan of 0.125% yielding consisted of rhamonse, arabinose, mannose, galactose and glucose with an estimated molecular weight of 1.5×10 4. ②Oral administration of coixan to normal mice showed no obvious action to decrease blood glucose level. Peritoneal administration of coixan in the dosage from 50 to 100 mg/kg was able to decrease blood glucose level significantly in normal mice, diabetic and hyperglycemic mice ( P <0.05 to P <0.01) in dosage dependent manner. Conclusion Coixan is able to decrease the blood glucose level in normal, diabetic and hyperglycemic mice and its hypoglycemic action is determined by the route of administration.
