摘要
目的:通过测定育龄期不孕妇女卵泡早期、采卵日、黄体中期血清白血病抑制因子(LIF)的含量,阐明LIF在血清中的变化规律,探讨血清中LIF与妊娠结局的关系,揭示LIF在胚胎种植过程中的可能作用机制,进而为提高人类辅助生殖技术的胚胎种植率和临床妊娠率提供理论依据。方法:选择在宁夏医科大学附属医院生殖医学中心首次实施体外受精-胚胎移植(IVF-ET)/卵泡浆内单精子显微注射(ICSI)治疗的不孕症妇女48例,采集同一患者自然周期卵泡早期、黄体中期血清以及IVF-ET周期中采卵日的血清及卵泡液,应用ELISA法测定血清及卵泡液中LIF的含量。结果:卵泡早期、采卵日、黄体中期血清LIF含量比较差异有统计学意义(P<0.05),采卵日血清LIF含量明显高于卵泡早期和黄体中期,黄体中期血清LIF含量略高于卵泡早期;妊娠组采卵日血清LIF含量比卵泡早期血清LIF含量的增加值明显高于非妊娠组(P<0.05)。结论:卵泡早期、采卵日、黄体中期血清LIF含量比较差异有统计学意义,提示月经周期中血清LIF来源的部位随时相而异;IVF-ET周期中采卵日血清LIF主要来源于卵巢;卵泡早期血清LIF及采卵日血清LIF的增加幅度可能预测妊娠结局。
Objective:To explore the change regularity of serum leukemia inhibitory factor(LIF) by detecting the contents of LIF during early follicular phase,on the day of ovum pick-up(OPU),and during midluteal phase in infertile women of childbearing age,explore the relationship between serum LIF and pregnancy outcome,and reveal the probable mechanism of LIF during embryo implantation,in order to provide a theoretical basis for improving embryo implantation rate and clinical pregnancy rate of human assisted reproductive technology.Methods:Forty-eight infertile women who received IVF-ET/ICSI in the hospital for the first time were selected,the serum samples and follicular fluid samples during early follicular phase,during midluteal phase,and on OPU day of IVF-ET were collected,ELISA was used to detect the contents of LIF in serum and follicular fluid.Results:There was statistically significant difference in serum LIF content among early follicular phase,on OPU day of IVF-ET,and during midluteal phase(P0.05),serum LIF content on OPU day of IVF-ET was higher than those during early follicular phase and midluteal phase,serum LIF content during midluteal phase was slightly higher than that during early follicular phase;the increasing amplitude of serum LIF content from OPU day of IVF-ET to early follicular phase in pregnancy group was higher than that in non-pregnancy group(P0.05).Conclusion:There was statistically significant difference in serum LIF content among during early follicular phase,on OPU day of IVF-ET,and during midluteal phase,which indicates that the source sites of serum LIF during menstrual cycle varies with time;serum LIF on OPU day of IVF-ET is mainly from ovary;the increasing amplitudes of serum LIF during early follicular phase and on OPU day of IVF-ET may be helpful to predict pregnancy outcome.
