阿霉素心肌病大鼠心钠素与丙二醛和超氧化物歧化酶及心室重构的关系 被引量:3

Relationship between atrial natriuretic peptide with malondialdehyde,superoxide dismutase and ventricular remodeling in rats with adriamycin cardiomyopathy

摘要 目的探讨阿霉素心肌病(ADR-DCM)大鼠心钠素(ANP)与丙二醛(MDA)、超氧化物歧化酶(SOD)及心室重构的关系。方法以腹腔注射盐酸阿霉素的方法建立大鼠ADR-DCM模型,另设正常对照组;用放射免疫法测定血浆及心肌(心房)匀浆上清液中ANP含量,测定左室质量指数(LVMI),经颈动脉测量左室收缩压(LVSP)、左室舒张末压(LVEDP)、左室压力最大上升及下降速度(±LVdp/dtmax)等心功能指标,测定心肌(左室)匀浆上清液中MDA、SOD含量;病理学切片检测,HE染色,显微镜观察。结果 ADR-DCM组大鼠血浆及心肌(心房)ANP含量显著升高(P<0.01),LVMI显著增加(P<0.01),LVSP、±LVdp/dtmax下降(P<0.01),LVEDP升高(P<0.01),心肌(心房)MDA含量升高(P<0.01),SOD含量降低(P<0.01)。ADR-DCM大鼠血浆ANP含量与±LVdp/dtmax呈负相关(r=-0.78,P<0.05);与LVMI呈正相关(r=0.80,P<0.01)。结论阿霉素通过氧化应激机制损伤心肌,从而导致心室重构及心功能不全,ANP能够反映其中氧化应激及心室重构的程度。 Objective To investigate the relationship between atrial natriuretic peptide (ANP),malondialdehyde (MDA), superoxide dismutase (SOD) and ventricular remodeling of rats with adriamycin eardiomyopathy. Methods To establish ADR induced dilated cardiomyopathy (DCM) model of rats by the method of iutraperitoneal injection of doxorubiein hydrochloride (ADR), and in addition the control group was set up. To determinate plasma ANP by radioimmunoassay (RIA) and left ventricular mass index (LVMI). The cardiac function indexes including left ventricular systolic blood pressure (LVSP), left ventrieular end-diastolic pressure (LVEDP), and the largest increased rate and declined of left ventricular pressure (±LVdp/dt) were determinated via the carotid artery. The MDA and SOD content in homogenate supernatant of left ventricle (LV) were detected.The patho- logical sections detection, HE staining and microscopy were performed. And ANP in homogenate supernatant of atrium was de- termined by RIA. Results In DCM rats, plasma and myocardial (atrium) ANP was significantly increased (P〈0.01), LVMI was increased (P〈0.01), LVSP and ± LVdP/dtmax were declined (P〈0.01), and LVEDP was increased (P〈0.01), myocardial(atrium) MDA was increased(P〈0.01 ) ,SOD activity was decreased(P〈0.01 ). Plasma ANP content and the cardiac function (±LVdp/dtmax) were negatively correlated in ADR-DCM rats (r=-0.78 ,P〈0.05 ). Plasma ANP content was positively correlated with LVMI in ADR-DCM rats(r=0.80 ,P〈0.01 ). Conclusion Doxorubicin injures myocardium through oxidative stress mechanism and leads to ventricu- lar remodeling and cardiac dysfunction. ANP may reflect the degree of oxidative stress and ventricular remodeling.
