摘要
研究影响佛手瓜SRAP扩增反应的主要因子,建立佛手瓜SRAP反应的优化体系,利用SRAP技术对福建省10个佛手瓜栽培种进行亲缘关系分析.筛选出10对具有较高扩增多态性的引物组合,共扩增出条带114条,其中62条为多态性条带,多态率为54.38%.聚类分析结果表明,当遗传距离为0.20时,可将10个品种分为3个类群;遗传距离为0.35时,10个品种聚为一类,表明10个佛手瓜品种的亲缘关系较近.
The main factors affecting the amplification results in the reaction system of sequence-related amplified polymorphism (SRAP) were investigated. SRAP-PCR reaction system of Seehium edu/e was optimized and SRAP was assessed to analyze the ge- netic relationships among 10 Sechium edule cultivars. A total of 114 DNA bands were amplified by 10 selective primers, 62 of which (54.38%) were polymorphic. Cluster analysis showed that the 10 cultivars were dassified into three duster groups with the genetic similarity of 0.20. All cultivars clustered together when the genetic distance was 0.35, which suggested the closer genetic relation- ship of 10 Sechium edule cultivars.
