摘要
肿瘤的多药耐药(MDR)是导致肿瘤难以根治、化疗失败和疾病复发的主要原因之一。肿瘤干细胞(CSC)是驱动肿瘤发生、演进、转移和复发的根源,是造成肿瘤耐药的根本原因。CSC主要耐药机制包括:ABC转运体增强CSC的抗药性、高水平表达抗凋亡基因、DNA修复能力增强以及低氧龛环境等。研究CSC生物学特性,阐明其耐药的分子机制,制定出针对CSC的靶向治疗方案以克服和逆转CSC的多药耐药,有助于提高肿瘤的治愈率和降低复发率。
Multidrug resistance (MDR) is the main obstacle for cancer to cure, failure of chemo- therapy and relapse. Cancer stem cells (CSCs) are driven force for tumorigenesis, the evolution of metastasis and recurrence, and the most fundamental reason for cancer drug-resistance. The primary drug resistance mech- anism includes : ABC transporters strengthening CSCs drug resistance, high level expression of the anti-apoptot- ic genes, DNA repair capacity enhancement and hypoxic niches, and so on. To study CSCs biological charac- teristics and clarify the resistance molecular mechanisms, and to develop a targeted therapy for CSCs, and then to overcome and reverse the multidrug resistance of CSC, will enhance cancer cure rate and reduce the relapse rate.
