双重氧化脱黑色素法在恶性黑色素瘤免疫组化诊断中的应用 被引量:11

摘要 目的:探讨双重氧化脱黑色素法在恶性黑色素瘤免疫组化诊断中的应用。方法:20例经病理确诊为恶性黑色素瘤的石蜡组织标本,用5种不同浓度的过氧化氢液先氧化脱色,经观察选取15%的过氧化氢液氧化30min后,再用0.5%高锰酸钾液加强氧化5min,最后5%草酸液清洁5min,应用Envision免疫组化法检测Melan-A、Melanoma、S-100和VIM阳性的表达。结果:20例恶性黑色素瘤切片在脱色素前HE染色可见棕黄色色素颗粒,经双重氧化脱色素处理后,HE染色效果好,无脱片;应用15%的过氧化氢脱色效果优于其他4种浓度的过氧化氢,滴加0.5%高锰酸钾5min,再滴加5%草酸液5min,黑色素完全消失;脱色处理后Melan-A、Melanoma、S-100和VIM阳性定位准确,着色好。结论:采用双重氧化脱黑色素法进行免疫组化染色具有较理想的作用,值得采用。 OBJECTIVE: To investigate the removal method of double oxidation of melanin in immunohistoehemical diagnosis of malignant melanoma. METHODS: The paraffin samples from 20 patients diagnosed as malignant melanoma were treated with five different concentrations of hydrogen peroxide bleaching solution before oxidation, After 30 minutes 15 % hydrogen peroxide solution were selected and treated with 0.5 % potassium permanganate solution to enhance oxida tion of 5 minutes and 5 % oxalic acid cleaning solution was used for 5 minutes,Envision immunohistochemical assay was used to detect Melan-A, Melanoma, S-100andVIM expression. RESULTS: The demelanin efficacy was better in 15 % of the oxidation of hydrogen peroxide solution for 30 minutes than other concentrations, Adding 0.5 % potassium permanganate solution for 5 min and 5 % oxalic acid for 5 rain could enhance oxidation and completely remove the melanin. CONCLU- SION: Double oxidation of de-melanin in immunohistochemical staining is a recommendable method in diagnosis of mela noma.
