摘要
目的探讨丹参酮ⅡA对大鼠失神经腓肠肌肌萎缩的防治作用。方法建立大鼠腓肠肌失神经支配模型,共48只,随机分为两组,丹参酮组:术后每天腹腔注射丹参酮ⅡA磺酸钠注射液;对照组:注射等体积生理盐水。术后4、6周每组随机取6只检测肌湿重,行HE和天狼星红染色,测量肌纤维横截面积、胶原纤维含量,另6只予以单宁酸—氯化铁媒染腓肠肌血管,测量毛细血管密度。结果随失神经时间延长两组均有肌纤维萎缩变细,肌纤维横截面积减小,胶原纤维增生;但术后4、6周丹参酮组肌湿重均重于对照组(P<0.05);肌纤维横截面积、微血管密度明显高于对照组(P<0.05),胶原纤维量明显少于对照组(P<0.05)。结论丹参酮ⅡA具有明显延缓大鼠腓肠肌失神经肌萎缩的作用,与其改善微循环,抑制胶原纤维增生有关。
Objective To explore the prevention and treatment effect of tanshinon RA on denervat- ed gastrocnemius muscular atrophy in rats. Methods The models of denervated gastrocnemius muscular at- rophy were prepared, totally 48 rats. They were randomized into two groups. In a tanshinon group, the intraper- itoneal injection of tanshinon IlA sodium sulfonate was applied every day after operation. In a control group, the physiological saline of the same volume was injected. 6 rats were randomized in 4 and 6 weeks after opera- tion for the detection of muscle humid weight. HE and Sirius red staining was adopted for the measuring of the cross sectional area of muscular fiber and the content of collagen fiber. Additionally, tannin - ferric chloride medium was used to stain gastrocnemius blood vessel for the measuring of capillary density. Results With the time passing of denervation, the muscular atrophy and thinning occurred, the cross sectional area of mus- cular fiber became smaller and the collagen fiber was of hyperplasia in both groups. In 4 and 6 weeks after operation,the muscle humid weight in the tanshinon group was heavier than that in the control group (P 〈 0.05 ) , the cross sectional area of muscular fiber was bigger and microvascular density was higher than those in the control group ( P 〈 0.05 ), the content of collagen fiber was less apparently than that in the control group(P 〈 0.05 ). Conclusion Tanshinon ⅡA delays denervated gastrocnemius muscular atrophy of the rats, which is related with the improvement of microcirculation and inhibition of collagen fiber hyperplasia.
