摘要
目的 探讨重组粒细胞集落刺激因子 (rhG CSF ,G CSF)对脐血CD3 AK细胞及其培养上清液诱导K562 细胞凋亡的影响。方法 用细胞形态学观察 ,DNA琼脂糖电泳 ,原位末端标记法对K562 ;CD3 AK细胞诱导的K562 ;CD3 AK细胞与大小剂量的G CSF共同诱导的K562 ;CD3 AK细胞培养上清液 (culturalsupernatants,CS)诱导的K562 ;CS与大小剂量的G CSF共同诱导的K562 进行检测分析。结果K562 自然凋亡率为 (3.5± 1.0 ) % ;CD3 AK细胞诱导的K562 凋亡率为 (2 7.4± 4.9) % ;CD3 AK细胞与大小剂量G CSF共同诱导的K562 细胞凋亡率分别为 (7.4± 3.0 ) %、(12 .9± 3.1) % (F =10 7.94,P <0 .0 1)。CS诱导的K562 细胞凋亡率为 (33.1± 5 .2 ) % ;CS与大小剂量的G CSF共同诱导的K562 细胞凋亡率分别为 (6 .4± 2 .3) % ;(14.8± 2 .5 ) % (F =183.36 ,P <0 .0 1)。结论 G CSF能部分或全部逆转脐血CD3 AK细胞和其培养上清液诱导的K562 细胞的凋亡。
Objective To explore the effect of G CSF on the K 562 cell apoptosis induced by cord blood CD 3AK cells and supernatants. Methods Apoptosis was detected by techniques of TDT end labelling, cell morphology and DNA agarose gel electrophoresis. The percentage of apoptosis was determined in K 562 cells, K 562 cells induced by CD 3AK cells, K 562 cells induced by both CD 3AK cells and G CSF, K 562 cells induced by the supernatants of cultured CD 3AK cells, as well as K 562 cells induced by both the supernatants and G CSF.Results The percentages of K 562 cell apoptosis in the corresponding tuations mentioned above were (3.5±1.0)%, (27.4±4.9)%,(12.9±3.1)%, (7.4±3.0)%,(33.1±5.2)%,(14.8±2.5)% and(6.4±2.3)%, respectively( F =107.94, P <0.01; F =183.36, P <0.01). Conclusion G CSF could partially or totally inhibit K 562 cell apoptosis induced by cord blood CD 3AK cells and the supernatants.
