利用木薯酒糟液培养枯草芽孢杆菌的工艺优化 被引量:2

摘要 以木薯酒糟液作为芽孢杆菌基础发酵培养基,筛选适合液体发酵的芽孢杆菌菌种,并通过单因子和正交试验方法,对木薯酒糟液培养芽孢杆菌的培养基配方及发酵条件进行了优化。结果表明,水产养殖用枯草芽孢杆菌在木薯酒糟液发酵培养基中的活菌数最高,达14.3×108cfu/mL;以该菌株作发酵菌种,得到最优培养基配方为:木薯酒糟液96.6%,葡萄糖2.0%,复合氮源营养液0.10%,尿素0.30%,蔗糖1.0%。最优培养条件为:pH值为6.0,温度37℃,摇床170r/min,发酵培养24h。采用优化后的木薯酒糟液发酵培养基和发酵工艺,发酵结束后培养液中水产养殖用枯草芽孢杆菌活菌数达35.0×108cfu/mL,分别是木薯酒糟基础发酵培养基和优化前的木薯酒糟发酵培养基中活菌数的9.46倍和2.45倍,发酵菌液经离心、干燥后制备的固体菌剂活菌数达140×108cfu/g。 The cassava lees waste water was used as basic fermentation medium for Bacillus. The Bacillus strains, which grows well in liquid fermentation, were screened. Then, the medium formula and fermentation conditions were optimized through single factor test and orthogonal test. The results showed that the viable count of Bacillus subtilis for aquaculture was highest in cassava lees wastewater medium and reached 14.3 ×10^8cfu/mL. The optimal medium composition for this strain was determined as: cassava lees wastewater 96.6%, glucose 2.0%, nitrogen solution 0.10%, urea 0.30% and sucrose 1.0%. The optimal culture conditions were: pH 6.0, temperature 37℃, shaking speed 170r/min and culture time 24 h. After optimization, the viable count ofB. subtilis for aquaculture reached 35.0×10^8cfu/mL. After being centrifuged and dried, the dried B. subtilis was prepared with a viable count of 140×10^8cfu/g.
