摘要
目的观察六味地黄汤及其水提醇沉物对2型糖尿病肾阴虚证大鼠脂代谢的影响。方法采用高脂乳灌胃结合腹腔注射小剂量链脲佐菌素(STZ)再灌胃氢化可的松的方法建立2型糖尿病肾阴虚证大鼠模型。设空白组、模型组、六味地黄汤组、六味地黄汤水提醇沉物组、罗格列酮组,分组给药30 d,模型组和空白组给等量生理盐水,检测各组大鼠空腹血糖(FBG)、血清cAMP、胆固醇(CHO)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDLC)、高密度脂蛋白胆固醇(HDLC)。结果模型大鼠FBG≥16.7 mmoL/L,cAMP水平为(1.32±0.04)mmoL/L,成功建立了2型糖尿病肾阴虚证大鼠模型;六味地黄汤及其水提醇沉物均能降低2型糖尿病肾阴虚证大鼠空腹血糖值及CHO、TG、LDL-C水平,升高HDLC水平,与模型组比较差异有统计学意义(P<0.01或P<0.05),改善脂代谢。结论六味地黄汤干预2型糖尿病肾阴虚证的作用可能与其改善脂代谢有关,其水提醇沉物可能是六味地黄汤改善2型糖尿病肾阴虚证的药效物质基础之一。
Objective To observe the effcts of Liuwei Dihuang Decoctionand its glycosides parts on lipid metabolism of the rat model of type 2 diabetic kidney yin deficiency syndrome. Methods Used the method of gavage high- fat milk, intraperitoneal injection of small doses of streptozotocin and gavage the Hydrocortisone to establish the rat model of type 2 diabetic kidney yin deficiency syndrome. All rats were divided averagely into normal group, model group, Liuwei Dihuang Decoctiongroup, Liuwei Dihuang Tang' s glycosides parts group and Rosiglitazone group, Respectively gavage drugs or Physiological saline 30 days, determine cAMP levels with ELISA, and detect the level of FBG, CHO, TG, LDL-C, HDL-C in each group. Results The FBG of model group≥ 16.7 mmoL/L (P〈0.01) , the value of cAMP is 1.32±0.04 mmoL/L (P〈0.01 ) , Successfully established the rat model of type 2 diabetic kidney yin deficiency syndrome with high- fat milk, small doses of streptozotocin and hydrocortisone; Liuwei Dihuang Decoctionand its glycosides parts are able to reduced the level of FBG, CHO, TG, LDL-C, and increased HDL-C (P〈0.01 或P〈0.05 ) , Improve the lipid metabolism. Conclusion The intervention effect of Liuwei Dihuang Decoctionon type 2 diabetic kidney yin deficiency syndrome may related with its improvement of lipid metabolism, and its glycosides parts may be one of effective substance basis of Liuwei Dihuang Decoctionimproving the type 2 diabetic kidney yin deficiency syndrome.
