摘要
目的探讨黄芪多糖(astragalan,AG)预处理对大鼠全脑缺血再灌注后认知功能的影响及其可能机制。方法 60只SD雄性大鼠随机分为假手术组、模型组及黄芪多糖预处理组。黄芪多糖预处理组每日腹腔注射黄芪多糖30mg/(kg·d),连续7d,最后一次给药后30min建立急性全脑缺血10min再灌注模型。模型采用四血管阻断全脑缺血模型。再灌注后24h每组各取5只,右侧大脑采用干湿重法计算脑水含量,左侧大脑取海马检测髓过氧化物酶(MPO)活力和NF-κB的DNA结合活力。其余每组各15只大鼠分别于再灌注前、再灌注后6h、12h、24h、48h、96h进行改良NSS评分;再灌注后第五日开始进行Morris水迷宫实验。结果模型组脑水含量、MPO活力、NF-κB的DNA结合活力、NSS评分、平均潜伏期及探索实验中在第二象限时间比与假手术组比较差异有统计学意义(P<0.05);黄芪预处理组与模型组比较,脑水含量降低,MPO活力和NF-κB的DNA结合活力下降,NSS评分降低,平均潜伏期缩短,第二象限时间比增高(P<0.05)。结论黄芪注射液可减轻大鼠全脑缺血再灌注后脑水肿程度,减轻海马区炎症反应,改善认知功能。
OBJECTIVE To study the effect of astragalan preconditioning on cognitive function in rats with global cerebral ischemia-reperfusion injury so as to investigate its probable mechanism.METHODS Sixty SD male rats were randomly divided into sham operated group,model group,and astragalan preconditioning group(30mg/(kg·d)).The animals were intraperitoneally administered with drugs for 7 days.Thirty minutes after the last administration,acute injury models with 10-minute ischemia and persistent reperfusion were blocked with Pullsinelli-4 vessel occlusion.Five rats in each group were killed to obtain their brains 24h after successful modeling.The water content in the right brain were measured by dry-wet method and the activity of MPO and NF-кB in the left brain were measured.Then neurological severity scores(NSS) of the other 15 rats in each group were observed respectively before reperfusion and 6h,12h,24h,48h,96h after reperfusion.Morris water maze test was employed to assess spatial learning and memory respectively five days after reperfusion.RESULTS The above mentioned indexes in the model group showed differences compared with those in the sham operated group(P0.05).Compared to the model group,the water content,the activity of MPO and NF-кB in the brain tissue,the NSS,the mean escape latency decreased,and the percent of time course in the second quadrant increased in astragalan preconditioning group(P0.05).CONCLUSION Astragalan could reduce the cerebral edema degree,relieve the abnormity of inflammatory reaction,exert protective effect on cognitive impairment in global cerebral ischemia reperfusion injury rats.
