舒血宁对高氧暴露早产鼠视网膜HIF-1α和VEGF表达的影响

The intervention effect of Shuxuening on the expression of HIF-1α and VEGF of retino in premature rat modle with hyperoxia--induced retinopathy

摘要 目的;观察舒血宁对高氧暴露早产鼠视网膜缺氧诱导因子-1α(hypoxia inducible factors-1α,HIF-1α)和血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影嗡。方法:3日龄早产大鼠36只随机分为:高氧组、干预组、空气组,免疫组织化学法检测各组早产鼠视网膜HlF-1和VEGF表迭水平。结果:空气组HIF-1α、VEGF均呈低表达,未见或罕见突破内界膜的内皮细胞核;与空气组相比,高氧组突破内界膜的内皮细胞械明显增多,HIF—1α及VEGF表达的表达均显著增加,(P分别〈0.001,0.001,0.01),与高氧组相比,干预组突破内界膜的内皮细胞撮明显减少,HIF-1α及VEGF的表达均显著下降(P分别〈0.05,0.001,0.05);视网膜HIF-1a的表达与VEGF的表达、视网膜HIF-1α的表达与突破税、网膜内界膜内皮钿胞棱数、VEGF的表达与突破视网膜内界膜内皮细胞核数均呈正相关(r分别为0.55,0.65,0.46,P分别〈0.01,0.01,0.05)。结论:高氧暴露早产鼠视网膜内可观察到VEGF和HIF--1α的高表达,舒血宁使高氧暴露早产鼠视网膜HIF-1和vEGF的表迭明显下降,突破视网膜内界膜的内皮细胞棱数量明显减少,减少新生血管的形成,对高氧性视网膜病有-定的保护作用。 Objective, To investigate the effects of Shuxuening in premature rat modle with hyperoxia-- induced retinopathy. Methods: Thirty-- six premature Sprague--Dawley rats were randomly divided into into 3 groups, hyperxia group, Shuxuening treatment group and air group, hyperxia group and Shuxuening treatment group were exposed to ≥90% O2 for 7 days,then exposed to air for 5 days,air group exposed to room air. Shuxuening treatment group were intraperitoneal injeetied of Shuxuening 1.0mg/kg, hyperxia group and air group were intraperitoneal injectied same dose of 0. 9% sodium chloride. Expressions of hypoxia inducible factors--1≥(HIF-1α) and vascular endothelial growth factor(VEGF)were examined with immunohistochemical staining, The numbers of endotheliocyte nuclei of neovaseularization which broke through the retlnal inner limiting memberane were recorded. Result:In air group, the numbers of endotheliocyte nuclei of neovascularization which broke through the retinal inner limiting memberane were not found or seldem found,expressions of HIF--1α and VEGF were low level ; while compared with the air group, all the three above of hyperxia group all greatly increased, P〈0. 001,0. 001,0.01, respectively. Compared with the hyperoxia group, all the three above of Shuxuening treatment group all greatly decreased, P〈0.05,0.001,0. D5, respectively. , there are positive correlation between H IF-1α,and VEGF , HIF-1αand the numbers of endotheliocyte nuclei-of neovaseularization which broke through the retinal inner limiting memberane, VEGF and the numbers of endotheliocyte nuclei of neovascularization which broke through the retinal inner limiting memherane, r= 0.55 ,0.65,0.46,P 〈0. 01,0. 01,0. 05, respectively, Conclnsion: Shuxuening have protective effects in hyperoxia--induced retinopathy in premature rat.
