摘要
目的:探讨白花香莲解毒方抗大鼠肝纤维化的作用及机制。方法:SD大鼠60只,随机分为空白组、模型组、秋水仙碱组、白花香莲解毒方组,采用四氯化碳(CCl4)皮下注射及高脂低蛋白饮食复合因素诱导复制大鼠肝纤维化模型,采用自动生化仪检测各组血清ALT、AST、ALB,ELISA法检测血清HA、LN及PC-Ⅲ的含量,HE染色法观察肝脏病理学变化,免疫组化法检测肝组织中转化生长因子β1(TGF-β1)的表达。结果:白花香莲解毒方能降低血清ALT、AST水平,增加ALB含量,减少HA、LN及PC-Ⅲ含量,改善肝组织纤维化程度,下调肝组织中TGF-β1表达,与模型组、秋水仙碱组比较,差异有统计学意义(P<0.05)。结论:白花香莲解毒方具有抗肝纤维化作用,其机制可能与干预TGF-β1表达有关。
Objective： To investigate the anti-hepatic fibrosis mechanism of baihua xianglian detoxifieation formula in Zhuang folk medicine. Methods：Sixty SD rats were randomly divided into four groups, blank group, model group, colchicine group, and baihua xianglian detoxification formula group. Subcutaneous injection of carbon tetrachloride was applied along with diet of high fat and low protein was given in order to induce model of hepatic fibrosis in rats. Besides, serum ALT, AST and ALB in all groups were detected through automatic biochemical analyzer. Serum HA, LN and PC-Ⅲ were measured with ELISA. Pathological changes of liver were observed under HE staining method. TGF-β1 in liver tissue was detected by immunohistochemical method. Results：Baihua xianglian detoxification formula could decrease serum ALT, AST, HA, LN and PC-HI, increase serum ALB, relieve hepatic fibrosis, and down-regulate expression of TGF-β1 in liver tissue, showing statistical differences when compared with model group and colchicine group （P 〈0.05）.Conelusion：Baihua xianglian detoxification formula in Zhuang folk medicine can relieve hepatic fibrosis, the mechanism of which may be related to intervention on expression of TGF-β1.
