RP-HPLC法同时测定石榴皮中4种多酚类成分的含量 被引量:14

Content Determination of 4 Polyphenols in Punica granatum by RP-HPLC

摘要 目的:建立同时检测石榴皮中没食子酸、石榴皮鞣素、安石榴苷和鞣花酸含量的方法。方法:采用反相高效液相色谱法。色谱柱为ArcusEP-C18(250mm×4.6mm,5μm),流动相为甲醇-0.1%三氟乙酸(梯度洗脱),流速为1.0ml/min,检测波长为254nm(没食子酸)、377nm(石榴皮鞣素、安石榴苷和鞣花酸)。结果:没食子酸、石榴皮鞣素、安石榴苷和鞣花酸的进样量分别在0.020~0.320、0.038~0.608、0.074~1.184、0.039~0.624μg范围内与各自峰面积积分值呈良好线性关系(r分别为0.9997、0.9971、0.9978、0.9994);平均加样回收率分别为93.22%、95.35%、98.00%、99.84%,RSD分别为2.75%、2.28%、2.11%、1.82%(n均为6)。结论:本方法精密、可靠,可作为检测石榴皮中没食子酸、石榴皮鞣素、安石榴苷和鞣花酸4种有效成分的方法。 OBJECTIVE:To develop a method for the content determination of gallic acid,punicalin,punicalagin and ellagic acid in Punica granatum.METHODS:RP-HPLC method was used.The determination was carried out on Arcus EP-C 18(250 mm× 4.6 mm,5 μm)with mobile phase consisted of methanol-0.1%TFA(gradient elution)at a flow rate of 1.0 ml/min.The detection wavelength of gallic acid was at 254 nm,and that of punicalin,punicalagin and ellagic acid were at 377 nm.RESULTS:The linear range was 0.020-0.320 μg for gallic acid(r=0.999 7),0.038-0.608 μg for punicalin(r=0.997 1),0.074-1.184 μg for punicalagin(r=0.997 8),0.039-0.624 μg for ellagic acid(r=0.999 4).The average recoveries were 93.22%(RSD=2.75%,n=6),95.35%(RSD=2.28%,n=6),98.00%(RSD=2.11%,n=6)and 99.84%(RSD=1.82%,n=6),respectively.CONCLUSION:The method is accurate,reliable and suitable for the content determination of gallic acid,punicalin,punicalagin and ellagic acid in P.granatum.
