反相高效液相色谱法同时测定西红花苷Ⅰ和西红花酸 被引量:6

Determination of Crocin Ⅰ and Crocetin Simultaneously by Reverse Phase High-Performance Liquid Chromatography

摘要 建立了反相高效液相色谱同时测定西红花苷Ⅰ和西红花酸的方法。采用C8色谱柱,流动相为甲醇水冰醋酸溶液,甲醇、水和冰醋酸比例为75∶24.5∶0.5,流速为0.6mL·min-1,检测波长为440nm,在该色谱条件下,西红花苷Ⅰ和西红花酸得到较好的分离,西红花苷Ⅰ和西红花酸浓度在0.625~40μg·mL-1范围内峰面积与进样量呈良好的线性关系,方法平均回收率为101.33%。该方法具有重复性好、准确、快速等优点,适用于西红花苷Ⅰ和西红花酸的同时测定。 In this study, we established a method for determination of crocin Ⅰ and crocetin simultaneously by re- verse phase High-Performance Liquid Chromatography. Elite C8 chromatographic column (4.6 mm ×250 mm, 5 μm) was used, the mobile phase was methanol water acetic acid solution, their proportion of methanol, water and acetic acid were 75: 24.5: 0.5, the detected wavelength was 440 nm, and the flow speed was 0.6 mL · min-1. The crocin Ⅰ and croeetin got better separation, both the contents of crocin Ⅰ and crocetin and their peak areas were in good linear relationships, respectively. The average recovery was 101.33%. This method showed good re- peatability, quantitative, accuracy and rapid, and could be used effectively for the simultaneous determination of crocin Ⅰ and crocetin.
