摘要
目的研究Sirt1和P55PIK在胃癌组织中的表达水平及其与临床病理参数的关系,并初步探讨Sirt1和P55PIK在胃癌组织中表达相关性。方法采用免疫组织化学方法检测45例胃癌组织及癌旁正常组织中Sirt1和P55PIK表达水平,同时提取15例胃癌及癌旁正常组织蛋白行Western blot检测Sirt1和P55PIK表达水平。结果 Sirt1和P55PIK在胃癌组织中的表达阳性率分别为55.56%和64.44%,明显高于癌旁组织中的24.44%和35.56%(P<0.05)。Sirt1和P55PIK表达与患者性别、年龄、肿瘤直径及浸润深度无明显相关性,但与胃癌TNM分期和淋巴结转移有关(P<0.05)。Western blot检测显示15例患者Sirt1和P55PIK在胃癌组织中表达水平明显高于癌旁正常组织。Spearman相关性分析显示,Sirt1与P55PIK在胃癌组织中表达呈正相关(r=0.363,P=0.015)。结论 Sirt1和P55PIK表达与胃癌淋巴结转移和肿瘤分期密切相关,表明在胃癌的发病过程中可能有Sirt1和P55PIK的参与,联合检测可作为判断胃癌预后和筛选高危转移患者的指标,并可作为有效的治疗靶点。
Objective To investigate the expression level of Sirt1 and P55PIK in the gastric cancer,and the correction between them.Methods Immunohistochemistry was used to detect the expression of Sirt1 and P55PIK mRNA and protein in 45 cases of gastric carcinoma and adjacent tissues.Western blot was used to detect the protein expression of Sirt1 and P55PIK in 15 cases of gastric carcinama and adiacent tissues.Results Positive rate of Sirt1 and P55PIK in gastric cancer was 55.56% and 64.44%,which was significantly higher than in the adjacent tissues(24.44% and 35.56%)(P0.05).The expression of Sirt1 and P55PIK in gastric cancer was not correlated with gender,age,tumor diameter and depth of invasion,but with TNM staging and lymph node metastasis(P0.05).Western blot revealed that the expression of Sirt1 and P55PIK in gastric carcinoma was higher than in the adjacent tissue.Spearman correlation analysis showed that Sirt1 and P55PIK in gastric cancer were positively correlated.Conclusion The expression of Sirt1 and P55PIK in gastric cancer is correlated with lymph node metastasis and tumor stage,suggesting their roles in the pathogenesis of gastric cancer.Combined detection of Sirt1 and P55PIK can be used to judge the prognosis of gastric cancer and screen the high risk of metastasis,and Sirt1 and P55PIK can serve as an effective therapeutic target.
