摘要
目的探讨新疆哈萨克族食管癌癌组织与癌旁正常组织间差异显著的DNA甲基化基因,初步建立哈萨克族食管癌患者特异的异常甲基化谱。方法采用Illumina公司Human Metylation 450K磁珠芯片,对哈萨克族食管癌癌组织、癌旁正常组织各6例共12个标本进行全基因组甲基化检测,分析二者之间差异显著的甲基化基因,对具有显著差异的甲基化基因进行基因类型(Gene Ontology,GO)分析。结果食管癌癌组织与癌旁正常组织间具有显著差异的DNA甲基化基因共40个,参与多个生物过程,如信号转导、细胞周期调控及细胞凋亡等。GO分析表明,基因CCND1,CCDC88C参与Wnt信号通路,DLL1参与Notch信号转导等。结论差异显著的甲基化基因为深入研究哈萨克族食管癌的发生机制提供了理论依据。
Objective To explore the methylation difference between cancer tissues and adjacent tissues of esophageal carcinoma (EC) ,and establish a preliminary methylation profile in Xinjiang Kazakh. Methods Six Kazakh esophageal carcinoma patients including 6 cancer tissues and 6 adjacent tissues were detected DNA methylation status using Illumina' s Infinium Methylation 450 K beadchip, to compare the difference and analyze the significant different genes online in GoMiner webside. Results Compared with adjacent tissues, 40 genes,which involved in many bioproeesses such as signal transduction, cell cycle, apoptosis et al, are significantly different in cancer tissues. CCNDI, CCDC88C were related to Writ signal pathway and DLL1 was related to Notch signal pathway by GO analysis. Conclusion Thesegenes will provide new targets for further more studies of EC.
