摘要 目的探讨TGF-β1和endoglin在乳腺癌发生发展过程中的表达及意义。方法免疫组化(S-P)方法,对30例乳腺正常组织、50例非典型性增生组织、50例原位癌组织和80例浸润性癌组织进行TGF-β1及endoglin蛋白的检测。采用RT-PCR方法检测endoglin mRNA的表达。结果 Endoglin蛋白标记的微血管密度值(MVD值)、endoglin mRNA以及TGF-β1蛋白在正常乳腺组、不典型增生组、原位癌组及浸润性癌组两两比较差异有统计学意义(P<0.05);不典型增生、原位癌及浸润性癌组织中TGF-β1蛋白表达阳性组中endoglin蛋白标记的MVD值及endoglin mRNA均显著高于TGF-β1表达阴性组(P<0.05)。结论 TGF-β1蛋白及endoglin mRNA和蛋白表达与肿瘤的进程密切相关,提示血管生成可能在乳腺癌发生发展过程中起重要作用,同时也提示TGF-β1可能对乳腺癌发生发展有促进作用,其对乳腺癌的早期诊断和判断预后有重要意义。 Objective To investigate the expression and significance of transforming growth factor (TGF-β1) and endoglin in breast cancer. Methods 210 samples including 30 cases of normal breast tissue (control group) ,50 cases of atypical hyperplasia tissues (atypical hyperplasia group) ,50 cases of in situ cancer tissues (in situ cancer group) and 80 cases of infiltrating carcinoma tissues (infiltrating carcinoma group) were collected from patients who underwent esophagectomy. The expressions of TGF-β1 and endoglin protein were detected by immunohistochemistry. The expression of endoglin mRNA was detected by RT-PCR. Results There were significant differences in the values of microvessel density (MVD) labeled by Endoglin protein, endoglin mRNA as well as TGF-β1 protein among control group, atypical hyperplasia group, in situ cancer group and infiltrating carcinoma group ( comparison between two groups, P 〈 0.05 ). The values of MVD labeled by Endoglin protein,endoglin mRNA in TGF-β1 protein positive expression groups in atypical hyperplasia tissues, in situ carcinoma tissues and infiltrating carcinoma tissues were significantly higher than those in TGF-β1 protein negative expression groups ( P 〈 0.05). Conclusion The expressions of TGF-β1 protein and endoglin mRNA as well as endoglin protein are closely correlated with the progress of carcinoma, which suggests that vascularization may play an important role in the pathogenesis and development of breast cancer, meanwhile, which suggests that TGF-β1 may have promotive effect on the pathogenesis and development of breast cancer, and TGF-β1 may play an important role in early diagnosis and prognosis judgment of breast cancer.
