摘要
目的研究复方丹参滴丸对过氧化氢损伤心肌细胞H9c2的保护作用及其对磷酸化CAMP反应元件结合蛋白(p-CREB)表达的影响。方法用0.3 mmol/L的H2O2处理心肌细胞H9c2,复制心肌细胞氧化损伤模型。将不同浓度的丹参滴丸(0.125 g/L、0.250 g/L和0.500 g/L)于H2O2处理前加入,分别用MTT法测定细胞生长抑制率,分光比色法检测乳酸脱氢酶(LDH)释放率,流式细胞仪检测细胞凋亡率,Western blot检测p-CREB表达水平。结果丹参滴丸可减轻由H2O2诱导的H9c2细胞凋亡和死亡,其生长抑制率与药物浓度呈依赖关系;丹参滴丸可以上调H9c2细胞中p-CREB的表达,下调LDH释放率(P均<0.05)。结论丹参滴丸可保护心肌细胞H9c2免受氧化损伤,并上调p-CREB的表达。
Objective To investigate the effect of composite salviae dropping pills on cardiomyocytes injure and p- CREB expression induced by H2O2. Methods H9c2 were injured by 0.3 mmol/L H2O2, different final concentrations of composite salviae dropping pills (0. 125, 0. 25 and 0.5 g/L) were added into H9c2 before injury. Cell viability was meas- ured by MTT assay. Cell apoptosis was assayed by flow cytometry (FCM). Expressions of p-CREB were detected by west- ern blot. Results H9c2 cardiomyocytes were injured by 0.3 mmol/L H2O2. Composite salviae dropping pills increased the cell viability apparently and inhibited the production of LDH induced by H2O2. Composite salviae dropping pills could effectively increase p-CREB expressions and inhibit cell apoptosis induced by H2O2 in H9e2 cardiomyocytes (all P 〈 0. 05 ). Conclusions Composite salviae dropping pills could up-regulate p-CREB expression and protect H9c2 from injury induced by H2O2.
