摘要
在长白山区选择具有代表性的56个野生北五味子自然群体,采用RAPD方法对其进行不同群体间遗传多样性分析。结果表明,在20个随机引物中共扩增出125条带(其中62个为多态性条带),平均每个引物扩增出6.25条带,多态率为49.6%;根据RAPD结果对其进行聚类分析的结果,群体间的遗传距离为0~0.56,显示出丰富的遗传变异。了解长白山区北五味子种质资源的遗传多样性将在保护和利用好种质资源、培育新品种等诸方面起到积极的作用。
RAPD marker was used to detect inter-population genetic diversity of S chinensis, fifty-six sam- ples, collected from typical populations, with using 20 random primers to investigate the genetic diversity among population. The 20 primers produced total of 125 bands (with 62 for polymorphism band), average of 6.25 bands per primer, and polymorphism rate was 49.6%. A dendrogram was prepared on the basis of a similarity matrix using the Unweighted Pair Group Method Arithmetic Mean (UPGMA) algorithm and the genetic distance ranged between 0 to 0.56, and revealed high genetic variation. An estimate of the ge- netic diversity and differentiation between genotypes of breeding germplasm is of key importance for its improvement. The results obtained from this study could be usefull in germplasm characterization and bet- ter plant breeding.
