亚急性瘤胃酸中毒时山羊下丘脑食欲调节因子mRNA的表达变化 被引量:1

摘要 研究干乳期山羊亚急性瘤胃酸中毒(SARA)时下丘脑相关食欲调节因子mRNA的表达变化。选择7只安装永久性瘤胃瘘管的健康干乳期山羊,随机分为对照组(3只)和试验组(4只),分别饲喂精粗饲料比为4∶6和6∶4的饲料,自由饮水,通过饲喂精粗饲料比6∶4饲料建立干乳期山羊亚急性瘤胃酸中毒模型。在采食前和采食后1~12 h收集瘤胃液测定其pH值和乳酸含量,于试验结束时宰杀动物采集下丘脑组织,液氮速冻后于-70℃保存待测。结果显示:(1)精粗饲料比6∶4日粮饲喂2周后成功诱导SARA状态(SARA组),采食后瘤胃液pH值低于5.8且持续时间约3 h,而精粗饲料比为4∶6组山羊瘤胃液pH均高于6.0(对照组),与对照组相比,SARA组山羊食欲显著降低。乳酸在采食后2~4 h下降,采食后6~8 h逐渐增加,但两组间无显著差异。(2)与对照组相比,SARA组刺鼠相关蛋白(AgRP)基因表达显著下调(P<0.05),神经肽Y(NPY)基因表达有下调的趋势,但SARA组山羊下丘脑中前阿片黑素细胞皮质激素(POMC)和促黑激素受体4(MCR4)的基因表达均无显著性变化(P>0.05)。从而提示:SARA时山羊厌食主要是由于下丘脑促食欲调节因子mRNA表达下调所致。
