摘要
目的:探讨CCK-8法和MTT法检测兔成纤维细胞活性的最佳实验条件,并比较其优略。方法:以传至第3代的兔成纤维细胞为研究对象,分别用CCK-8法和MTT法检测兔成纤维细胞的增殖活性。结果:MTT法的最佳检测波长490nm,CCK-8法最佳检测波长为450nm,两法的最佳检测时间均为加入试剂后4h。适宜检测的细胞数量范围为2×103/ml~1×105/ml个。结论:CCK-8法较MTT法检测的灵敏度高,准确性好,是一种优于MTT法的检测兔成纤维细胞增殖活性的方法。
Objective: To investigate optive experiment conditions between MTT and CCK-8 in Rabbit fibroblasts, and to compare which was better.Method: Rabbit fibroblasts of PC3 cell were studied.The activity of rabbit fibroblast was detected respectively with CCK-8 and MTT.Result: For MTT assay, the best wavelength was 490 nm; and the best wavelength of CCK-8 assay was 450 nm.The best test both were 4 h after adding reagent, the number of detect Rabbit fibroblasts of PC3 cell was within the range of 2 × 10^3/ml-1 x 105/mi.Conclusion: CCK-8 assay is better than MTT assay in the aspects of detection sensitivity and accuracy, and CCK-8 is superior to MTT in Rabbit fibroblasts proliferation assay.
