摘要
目的研究体外培养大鼠肠道菌及肝微粒体代谢酶对甘草主成分的代谢情况的影响。方法体外培养大鼠肠道菌和肝微粒体孵育体系,并对甘草提取物进行代谢研究。采用高效液相色谱法分析甘草提取物孵育前后主要成分的变化,并计算各代谢产物的代谢率。结果甘草中3种主要成分均可在大鼠肠菌液中产生明显的代谢作用,其中主成分1和主成分3(甘草酸)24h内代谢率接近100%,主成分2在24h内代谢率超过50%,而3个主要成分在大鼠肝微粒体中基本无代谢。结论体外培养大鼠肠道菌可有效转化甘草提取物主要成分,肝微粒体对甘草主成分的代谢作用不明显。
Objective To study the biological transformation function of rat intestinal flora and liver microsomal ma- tabolism enzyme on Liquorice Root extract in vitro. Method Liquorice Root extract was incubated with rat intestinal flo- ra culture medium and Liver microsomes incubation system in vitro. High performance liquid chromatography was applied to determine the contents of the main components of Liquorice Root extract before and after incubation. And the metabolic rates of main components were calculated. Result The 3 main components of Liquorice Root extract could be trans- formed in rat intestinal bacteria. Among them, the conversion rate of component 1 and glycyrrhizic acid was near 100% within 24 h, component 2 was more than 50%. Conclusion Rat intestinal bacteria incubated in vitro can transform the main components of Liquorice Root extract efficiently.
