摘要
目的:建立一种快速高效的豚鼠急性心房颤动(atrial fibrillation,AF)模型,探讨房颤对心房肌动作电位时程(APD)的影响。方法:高频刺激豚鼠右心房处诱发房颤,记录心电图(ECG)及单相动作电位(MAP)的改变。结果:6只豚鼠共给予Burst刺激70次,以AF诱发时长>1s为模型建立成功。实验共诱发AF成功62次,成功率为88.75%,平均时长为9.860±22.035s。模型建立成功的豚鼠心房率显著增加,由205.184±31.278次/min增加到661.312±251.401次/min(P<0.05)。心房肌APD10、APD20、APD50、APD90及P-P周期长度(CL)在AF发作时显著减小(P<0.05),分别由6.038±4.172ms、13.029±11.003ms、37.193±12.504ms、84.453±24.567ms、300.148±52.020ms减小到3.291±1.906ms、6.451±3.140ms、18.227±12.646ms、51.887±36.137ms、105.414±44.567ms。动作电位幅值由AF前的34.422±10.425mV降至AF时的15.285±7.499mV,显著降低(P<0.05)。结论:经高频刺激右心房方法制作房颤动物模型成功率高、重复性好,并伴有心房肌的电生理改变,是建立急性房颤模型的有效方法。
Objective:To establish an efficient acute atrial fibrillation(AF) model and observe the changes of action potential duration(APD) in AF.Methods:Atrial fibrillation was induced by rapid pacing the right atrium of guinea pigs,and the electrocardiogram(ECG) and monophasic action potential(MAP) was recorded.Results: 70 times rapid pacing in 6 guinea pigs were used as stimulation,and the durations of AF 1s were defined as success.AF models were induced 62 times through Burst stimulation,with 88.75% success.The guinea pig atrial rate increased significantly,from 205.184±31.278 times/min to 661.312±251.401 times/min(P〈0.05).During AF,the APD10,APD20,APD50,APD90 and cycle length(CL) significantly decreased(P0.05),and action potential amplitude significantly reduced(P〈0.05).Conclusion:The acutely rapid pacing – induced AF model is an effective method for the study of electrophysiological changes of atrial myocardium with high successful rate and repeatability.
