摘要
建立了高效液相色谱测定人血浆中愈创木酚甘油醚浓度的方法。采用Higgins C_(18)色谱柱(250mm×4.6mm 5μm),流动相为乙腈-0.1%三乙胺水溶液(pH3.0)(体积比27:73),流速1.0mL·min^(-1),内标为替硝唑,检测波长273nm。方法的线性范围为0.05-2.0μg·mL^(-1),r=0.9958,定量限为0.05μg·mL^(-1),平均提取回收率89.92%-97.13%,日内和日间RSD均小于10%。本方法灵敏、简便、准确、快速,符合生物样品分析要求,可用于人血浆中愈创木酚甘油醚浓度的测定和药代动力学研究。
A method for determining Guaifenesin concentration in human plasma was established by reversed phase high performance liquid chromatography(HPLC).The separation was achieved on a Higgins C_(18) column.The mobile phase was consisted of acetonitrile-0.1%triethylamine(pH3.0) (27:73,V/V) at a flow rate of 1.0mL ? min^(-1).The internal standard was tinidazole and the wavelength was 273nm.The linear range was 0.05—2.0μg ? mL^(-1),r = 0.9958.Lower limit of quantification was 0.05μg ? mL^(-1).The average extraction recovery was 89.92%to 97.13%and both intra-day and inter-day precisions were less than 10%.The established method is sensitive,simple,fast and accurate.This method is suitable for determination of Guaifenesin in human plasma.
