摘要
目的:探讨鸦胆子油乳对人宫颈鳞状上皮永生化细胞(Ect1/E6E7)体外增殖和凋亡的作用。方法:以不同浓度鸦胆子油乳(5、10、20、40、80μg/ml)作用于Ect1/E6E7细胞24、48与72 h,采用倒置显微镜观察细胞形态学改变,MTT法检测药物对细胞的体外增殖抑制作用。Hochest33258细胞核染色法检测不同浓度鸦胆子油乳(5、20μg/ml)体外作用于Ect1/E6E7细胞48 h后细胞凋亡形态学改变。流式细胞术Annexin V-FITC/PI法检测不同浓度鸦胆子油乳(5、10、20μg/ml)体外作用48h对Ect1/E6E7细胞凋亡率影响。结果:鸦胆子油乳对Ect1/E6E7细胞具有明显的抑制体外增殖作用,MTT结果显示抑制呈时间和浓度的依赖性(P<0.05)。经5、20μg/ml鸦胆子油乳体外作用48 h后,Hochest33258细胞核染色法显示细胞出现凋亡形态学改变,并产生凋亡小体。流式细胞仪检测显示,各实验组细胞经鸦胆子油乳作用48 h后,细胞凋亡率分别为(7.47±0.69)%、(10.93±1.15)%、(17.36±1.05)%,与对照组比较差异有统计学意义(P<0.05)。结论:鸦胆子油乳对Ect1/E6E7细胞有明显的增殖抑制作用,其机制可能与诱导和促进细胞凋亡相关。
Objective: To explore the effect of Brucea javanica oil emulsion ( B JOE) on in vitro proliferation and apoptosis of cer- vical immortalized cells (Ectl/E6E7) . Methods: Different concentrations of BJOE (5, 10, 20, 40, and 80 μg/ml) were used to treat Ectl/E6E7 cells for 24, 48, and 72 hours; inverted microscope was used to observe the changes of cell morphology; MTT method was used to detect the inhibiting effect of BJOE on in vitro proliferation of Ectl/E6E7 cells; Hochest 33258 nuclear staining method was used to detect the morphological change of apoptosis of Eetl/E6E7 cells at 48 hours after treated with different concentrations of BJOE ( 5, 10, and 20μg/ml) . Results: BJOE inhibited in vitro proliferation of Ectl/E6E7 cells significantly, the results of MTT method showed that the in- hibiting effect showed time - and dose - dependent manners ( P 〈 0. 05 ) ; at 48 hours after treated with 5 and 20μg/ml BJOE in vitro, the results of Hochest 33258 nuclear staining method showed that morphological changes of apoptosis occurred, and apoptosis body appeared ; the results of flow cytometry showed that at 48 hours after treated with BJOE, the cell apoptosis rates in different experimental groups were (7.47 ± 0. 69 ) % , ( 10. 93 ±1.15 ) %, and ( 17.36 ± 1.05 ) %, respectively, compared with control group, there was statistically signifi- cant difference ( P 〈 0.05 ) . Conclusion : B JOE has a significant inhibiting effect on proliferation of Ectl/E6E7 cells, and the mechanism may be correlated with inducing and promoting apoptosis.
