摘要
目的:探讨年龄对大鼠原代培养的阴茎海绵体平滑肌细胞(CCSMCs)分泌胰岛素样生长因子-1(IGF-1)水平的影响,及其与老年性ED的关系。方法:体外原代培养不同年龄段(4月龄、12月龄和24月龄)的大鼠CCSMCs,免疫组化染色鉴定;定量培养CCSMCs,采用酶免疫分析法(EIA)定量检测CCSMCs分泌IGF-1含量的变化,分析比较不同年龄对CCSMCs分泌IGF-1含量的影响。结果:成功原代培养CCSMCs,随着月龄的增加,CCSMCs分泌的IGF-1含量减少,与4月龄(7.1 ng/105个细胞)比较、12月龄(2.2 ng/105个细胞)和24月龄(1.9ng/105个细胞)CCSMCs分泌的IGF-1含量明显减少,有统计学意义(P<0.01)。结论:年龄是影响原代培养CCSMCs分泌IGF-1水平的因素之一,IGF-1的低表达可能和老年性ED的发生有关。
Objective： To observe the secretion of insulin-like growth factor-1 （IGF-1） in corpus cavemosum smooth muscle cells （CCSMCs） in rats of different ages and explore the possible relationship of IGF-1 with aging-related erectile dysfunction （ED）. Methods ： We primarily cultured CCSMCs of rats aged 4, 12 and 24 months, and identified them by immunohistochemistry. We quanti- tatively cultured the CCSMCs in 6-well culture plates, determined the levels of IGF-1 secreted from the CCSMCs by enzyme immunoas- say （EIA） and analyzed the effect of age on the IGF-1 level. Results： CCSMCs were successfully cultured in vitro. The level of IGF-1 secreted from the CCSMCs was decreased with the increase of age, with 7.1 ng/105 cells in the 4-month-old group, 2.2 ng/105 cells in the 12-month group, and 1.9 ng/105 cells in the 24-month group, with statistically significant differences among the three groups （ P 〈 0.01 ）. Conclusion ： The secretion of IGF-1 is reduced with the increase of age, and the decreased expression of IGF-1 might be associated with aging-related ED. Natl J Androl, 2013, 19 （1） ： 6 -9
