电针对帕金森病模型大鼠黑质细胞线粒体复合物活性的影响 被引量:11

摘要 目的通过检测帕金森病(PD)模型大鼠黑质细胞线粒体复合物活性的改变,探讨电针治疗对黑质细胞线粒体功能的影响,以期揭示电针治疗PD的机制。方法将48只SD雄性大鼠随机分成4组:正常组、溶剂对照组、模型组、电针治疗组;采用鱼藤酮颈背部皮下注射法制备模型,按行为学评分标准记分;治疗结束后采用免疫组织化学技术检测中脑黑质酪氨酸羟化酶(TH)阳性神经元数目;同时采用紫外分光光度法检测线粒体复合物Ⅰ~Ⅳ的活性。结果与模型组相比较,电针治疗组行为学异常明显改善(P<0.05)。电针治疗组黑质TH阳性神经元数目较模型组显著增加(P<0.01)。模型组的线粒体复合物Ⅰ的活性明显低于正常组(P<0.01),电针治疗组的线粒体复合物Ⅰ的活性明显高于模型组(P<0.01)。各组线粒体复合物Ⅱ、Ⅲ、Ⅳ无显著差异。结论粒体复合物Ⅰ功能障碍在PD发病中占有重要的地位,并可能是针刺治疗作用的靶点之一。 Objective To explore the effects of electroacupuncture treatment on nigral cell mitochondria function in Parkinson's disease (PD) model rats and reveal the mechanism of acupuncture treatment on PD by detecting mitochondrial complexes activity change of PD model rat nigral cell. Methods 48 SD rats were divided into 4 groups randomly: normal, solvent control, model and electroacupuncture therapy groups. PD model was made by subcutaneous injection of rotenone in the back of neck. The behavior of rat was scored according to the standard of the behavior science calculated. TH positive neuron count of substantia nigra was detected by immunohistochemieal method. The activity of mitochondrial complex I - IV were detected by ultraviolet spectrophotometry. Results Compared with model group, the abnormal changes of electroacupuncture therapy group were small(P 〈 0. 05 ). Compared with model group, TH positive neuron count of substantia nigra of electroacupuneture therapy group was increased significantly(P 〈 0. 01 ). The activity of mitochondrial complex I of model group was lower than that of normal group (P 〈 0. 01 ) , and the activity of mitochondrial complex I of electroacupuncture therapy group was lower than that of model group( P 〈 0. 01 ). While the activity of mitoehondrial complex II ,III and IV had no obvious difference. Conclusions Electroaeupuneture therapy can improve the abnormal behaviors and the activity of mitochondrial complexes and also can reduce the loss of dopamine neurons in substantia nigra. It explains that the dysfunction of mitochondrial I plays an important role in PD and it also can be one of the target points of electroacupuncture therapy.
