黄芩苷液体栓的制备及体外释放研究 被引量:2

Preparation of Baicalin-loaded liquid suppository and determination of the drug release in vitro

摘要 【目的】制备一种具有良好直肠应用前景的温度敏感性水凝胶(液体栓)。【方法】以黄芩苷(Baicalin,BC)为模型药物,以泊洛沙姆407(PoloxamerF127,P407)为主要凝胶基质,以冷法制备温敏凝胶,以相转变温度为主的多因素为指标,筛选出含药液体栓的最佳处方。进而借助透析袋-浆法以及紫外分光光度法,考察其体外释药性能。【结果】最佳处方为P407-RA-PEG400(18::05:1.0)。该处方,制成的胶凝温度约为35℃,7h内体外释药百分率达92%,含药温敏凝胶相比原料药不论在释药的速度和程度上均有显著优势,说明该剂型更能发挥黄芩苷的速释作用。【结论】制备出了一种更加安全、有效、稳定的载药温敏凝胶,以期为难溶性药物以及中药新剂型的开发做一点新的尝试。 【Objective】To prepare a thermosensitive in situ gel (liquid suppository) for rectal administration and develop a method for the determination of drug release.【Methods】The thermosensitive gel was prepared by Cold Method. The formulation screening was performed by taking sol-gel transition temperature and many other factors. UV and dialysis bag-pulp methods were used for the determination of drug release in vitro.【Results】The best formulation of BC thermosensitive gel was PoloxamerF127- SA-PEG4000 (18:0.5:1.0). The T gel of the best formulation was 35 ℃. In 7 hours, the drug release in vitro reached 92%. There were some distinct advantages of the thermosensitive in situ gel in the drug release of speed and extent. It indicated that the quick-release of BC was promoted by this type of formulation.【Conclusion】A drug loaded thermosensitive in situ gel is prepared, it is more safe, more effective and more stable.
