摘要
目的建立参芪养心滴丸中的哈巴俄苷及肉桂酸的含量测定方法。方法采用反向高效液相色谱法测定参芪养心滴丸中的哈巴俄苷及肉桂酸的含量,色谱条件为:Welchrom-C18色谱柱(200mm×4.6mm,5μm),以甲醇(A)-0.5%甲酸(B)为流动相进行梯度洗脱(0~40min,A∶B由50∶50~60∶40;40~45min,A∶B由60∶40~50∶50),流速为1mL/min,检测波长278nm,柱温25℃。结果哈巴俄苷在0.94~30.08μg/mL呈良好的线性关系(r=1.0),平均回收率为98.71%,方法精密度的RSD为0.92%(n=6);肉桂酸在0.44~14.0μg/mL呈良好的线性关系(r=1.0),平均回收率为100.84%,方法精密度的RSD为0.68%(n=6)。结论该方法简单、准确、重复性良好,可用于参芪养心滴丸的质量控制。
Objective To establish an accurate method for determination of Harpagoside and cinnamic acid in Shenqiyangxin drop pills. Methods Gradient HPLC method was set up to directly determine Harpagoside and cinnamic acid. The experiment conditions were as follows.. Kromasil-C18 (200 mm× 4.6 mm, 5 μm) column, the mobile phases consisted of methanol (A) and 0.5% acetic acid (B) using a gradient elution of 50%-40% (V/V) B at 0-40 min; 40%-50% B at 40-45 min. The flow rate was 1.0 mL/min and the detection wavelength was at 278 nm. Results There was a good linearity within the range of 0.94-30.08 μg/mL (r= 1.0) for Harpagoside and 0.44-14.0 μg/mL (r:0. 999 9) for cinnamic acid,the average recovery was 98.71% with RSD of 0.92% (n =6) for Harpagoside and 100.84% with RSD of 1.14% (n=6) for innamic acid. Conclusion The method is simple,accurate and reproducible. It can be used for the quality control of Shenqiyangxin drop pills.
