PI3K/AKT抑制剂对肾透明细胞癌中Notch1信号通路的影响 被引量:3

摘要 目的探讨PI3K/AKT信号通路在肾透明细胞癌中的活性及其抑制剂对Notch1信号通路的影响。方法收集36例肾透明细胞癌及其邻近非肿瘤肾组织,Western blot检测AKT(p-S473)和Notch1蛋白表达,分析AKT(p-S473)的表达与肿瘤临床特征之间的关系。PI3K/AKT抑制剂LY294002处理肾透明细胞癌细胞系786-O,Western blot和qrt-PCR检测HES1、Notch1蛋白及mRNA的变化,MTS法检测细胞增殖,流式细胞术检测其对细胞周期及凋亡的影响。结果肾透明细胞癌组织中AKT(p-S473)的表达明显高于癌旁组织,其表达与患者年龄、肿瘤大小、核分级及肿瘤TNM分期无明显关系(P>0.05)。PI3K/AKT抑制剂LY294002抑制786-O细胞增殖,促进细胞周期抑制,诱导细胞凋亡。抑制PI3K/AKT信号通路可降低HES1与Notch1蛋白表达。结论在肾透明细胞癌中,PI3K/AKT信号通路呈过度活化状态,并且与Notch1信号通路共同起促癌作用。PI3K抑制剂LY294002可以下调Notch1信号通路的活性,为肾透明细胞癌的治疗提供了新的治疗策略。
