转化生长因子-β1促进miR-181a表达诱导肝星状细胞-T6细胞活化 被引量:7

摘要 目的观察转化生长因子-β1(TGF-β1)对miR-181a表达的影响,以及miR-181a对体外培养的肝星状细胞(HSC)-T6细胞的作用。方法 TGF-β1处理HSC-T6细胞后,检测miR-181a和α-平滑肌肌动蛋白(α-SMA)表达。miR-181amimics转染HSC-T6细胞后,Western印迹检测α-SMA和Ⅰ型胶原a2链(collagenⅠa2,ColⅠA2)蛋白表达,CCK-8计数法检测细胞增殖。结果 TGF-β1处理组细胞中miR-181a和miR-181b表达量较对照组细胞明显升高,miR-181a转染组细胞中α-SMA和ColIA2表达量明显高于对照组细胞,miR-181a对HSC-T6增殖无明显影响。结论在HSC-T6中,TGF-β1促进miR-181a表达,miR-181a可促进HSC-T6细胞活化和分泌胶原,进一步明确了TGF-β1促进HSC-T6活化的机制。 Objective To observe the effects of TGF-β1 on miR-181a expression in HSC-T6 cells, and miR-181a on cultured HSC-T6 cells. Methods Real-time PCR was used to determine the expression of miR-181a after TGF-β1 induction, also the expression of a-SMA and Col I A2 protein, cell proliferation were determined to evaluate the effects of miR-181a on HSC-T6 cells. Results The expression of miR-181a was significantly increased after TGF-β1 induction. The expression of a-SMA and Col I A2 proteins were significantly increased after miR-181a mimics transfection, while cell proliferation was no effect. Conclusion Inducing miR-181a expression may be one of the mechanisms for TGF-β1 promoting HSC-T6 cells activation and secretion.
