银杏叶片高效液相色谱数字化定量指纹谱和对数指纹谱研究 被引量:2

Digitized and quantified fingerprints and logarithm fingerprints of Ginkgo tablets by high performance liquid chromatography

摘要 目的建立银杏叶片片(Ginkgo tablet,GT)数字化定量指纹图谱和对数指纹谱。方法采用RP-HPLC法,以Agilent Poroshell 120 SB C18色谱柱(4.6 mm×150 mm,2.7μm),0.2%磷酸水溶液-甲醇为流动相,线性梯度洗脱,流速为0.5 mL.min-1,柱温(35±0.15)℃,紫外检测波长为246 nm,进样量为5μL。应用46个数字化参数定量揭示GT-HPLC指纹图谱特征,用整体定量法鉴别14批次GT质量。结果以槲皮素(quercetin,QC)为参照物峰,确定37个共有指纹峰,建立了GT-HPLC数字化定量指纹图谱和对数指纹谱。用系统指纹定量法鉴定14批GT中质量极好者有4批,质量很好有1批,3批质量好,1批质量良好,质量为中者2批,质量为一般,差或劣的分别各1批。除个别批次外,对数指纹谱鉴定质量普遍升高。结论本法可清晰定量化揭示银杏叶片HPLC指纹谱超信息特征。对数指纹谱定量意义不显著,还可导致鉴定信息失真,但能消除批间差异和增加指纹共性特征,提供了一种新思路。 Objective To establish the HPLC digitized quantified fingerprints (FP) and logarithm fingerprint (LFP) of Gink- go tablets (GT). Methods The chromatographic fingerprints were developed by Agilent Poroshel[ 120 SB C18 (4.6mm ×150 mm,2.7μm) as the separation column and 0.2% mol ·L ^- 1 HaPOn-methanol as the mobile phase eluted with liner pro- gram. The flow rate was 0.5 mL · min^-1, the UV absorbance was monitored at 246 nm by injecting 5 μL of the sample solu- tion each time and the column temperature was maintained at (35± 0.15) ℃. Totally 46 digitized parameters were applied to evaluate the GT-HPLC fingerprints and entirely quantified method was used to identify the quality. Results The high perfor- mance liquid chromatography was employed to establish the digitized fingerprints amd 37 co-possessing peaks were marked with quercetin (QC) peak as the referential peak. The 46 digitized parameters were applied to evaluate the GT-HPLC FP and LFP and identify the quality of 14 batches of GTs by systematically quantified fingerprint method (SQFM). The quality of 4 batches was the best, 1 batch was better, 3 batches was good, and the others were all lower than average. However, except for very few batches, the identification results were generally increased by LFE Conclusion The method can quantitatively disclose the supper information in GT HPLC-FPs. LFP may get unreal results for GT quality. The differences between each batch can be eliminated through enhancing the common characteristics and provide a novel idea for the quality control of traditional Chinese medicine.
