摘要
目的探讨Twist和E-cadherin蛋白在大肠癌发生发展中的临床意义。方法采用免疫组化PV9000法检测78例大肠癌和42例大肠腺瘤组织中Twist和E-cadherin蛋白的表达。采用Westernblot法检测Twist蛋白在30例新鲜大肠腺癌组织和30例新鲜大肠腺瘤组织中的表达。结果Twist蛋白在大肠癌组织中的阳性率(56.4%)明显高于腺瘤组织(26.2%)和癌旁正常黏膜组织(10%);E-cadherin在大肠癌组织中的阳性率(48.7%)明显低于腺瘤组织(76.2%)和癌旁正常黏膜组织(100%)。Twist和E-cadherin在大肠癌中的异常表达均与肿瘤分化程度和淋巴结转移有关(P均<0.05),而与肿瘤大小无关(P>0.05)。E-cadherin的阳性表达与肿瘤的TNM分期有关(P<0.05),但Twist的阳性表达与TNM分期无关(P>0.05)。结论Twist和E-cadherin蛋白在大肠癌的发生及癌组织的侵袭和转移过程中可能起重要作用。
Objective To investigate the clinical significance of Twist and E-cadherin proteins expression in col-orectal carcinoma. Methods The expression of Twist and E-cadherin proteins was detected using immunohistochemistry in 78 cases of colorectal carcinoma and 42 cases of colorectal adenoma. Twist protein expression level was detected by Western blot method in 30 cases of colorectal carcinoma and 30 cases of colorectal adenoma. Results Higher positive rate of Twist was detected in colorectal carcinomas (56.4%) than that in colorectal adenomas (26.2%) and paracancerous normal tissues (10%). Lower positive rate of E-cadherin was detected in colorectal carcinomas (48.7%) than that in colorectal ade-nomas (76.2%) and paracancerous normal tissues (100%). The abnormal expression of Twist and E-cadherin were signifi-cantly correlated with the tumor differentiation and lymph node metastasis (P 〈 0.05), while not correlated with the size of tumor (P 〉 0.05). The abnormal expression of E-cadherin was significantly correlated with TNM stage (P 〈 0.05), but the abnormal expression of Twist was not correlated with it (P 〉 0.05). Conclusion Twist and E-cadherin are associated with tumorigenesis and play an important role in invasion and metastasis of colorectal carcinoma.
