泽兰乙醇提取物对大鼠血管的舒张作用及其机制研究 被引量:3

摘要 [目的]探讨泽兰乙醇提取物的舒张血管效应及其机制.[方法]利用多通道生理记录仪对大鼠离体胸主动脉进行观察,观察泽兰乙醇提取物对血管的作用,并从血管内皮细胞及一氧化氮(NO)/环磷酸鸟苷(cGMP)信号通路探讨其作用机制.[结果]泽兰乙醇提取物对内皮完整的大鼠胸主动脉环具有剂量依赖性舒张血管效应.去除血管内皮,可明显抑制泽兰的舒张血管效应;应用一氧化氮合酶抑制剂和鸟苷酸环化酶抑制剂预处理后,泽兰的舒张血管效应明显被阻断;应用盐酸普萘洛尔或阿托品对泽兰的舒张血管作用无影响.[结论]泽兰乙醇提取物的舒张血管效应存在内皮依赖性,且可能与NO/cGMP信号通路相关. OBJECTIVE To study the effects of ethanol extract of Lycopus lucidus Turcz (ELT) on vascular relaxation and its mechanism in rats. METHODS Using the multi polygraph, the effects of ELT on vascular relaxation were evaluated in organ bath, and its mechanisms were studied by the point of the vascular endothelium and NO/cGMP signaling pathway. RESULTS ELT had the effect of vasodilation with dose-dependent manner, and after removal of functional endothelium, ELT-induced vasodilation was abolishied. Pretreatment of the aortic rings with NG-nitro-L-arginine methyl ester (L-NAME) or 1H-J1, 2, 4]-oxadiazole-[4, 3-a]-quinixalin-l-one (ODQ) inhibited the relaxation induced by ELT. However, ELT-induced relaxation was not blocked by pretreatment of propranolol or atropine. CONCLUSION ELT has the effect of vasodilation, and its mechanism should be related with functional endothelium and NO/cGMP signaling pathway.
