摘要
目的:优化苜蓿黄酮的提取富集工艺。方法:采用均匀设计法对苜蓿黄酮提取工艺进行优化,采用大孔树脂对总黄酮进行富集工艺研究。结果:最佳提取工艺为固液比为1∶19,提取2次,提取时间128min,乙醇浓度71%,提取率预测值为34.2mg/g。大孔树脂富集纯化工艺采用D101树脂,以3BV用量的70%的乙醇作为洗脱剂进行洗脱,洗脱流速为2ml/min。结论:优化的苜蓿总黄酮提取、富集工艺经济、稳定、合理可行。
Objective:To optimize the extraction technology of total flavonoids from Medicago sativa.Methods:Select the contents of total flavonoids from extract solution as the target,the conditions of extraction were optimized by the uniform design method.Results:The optimum extraction technology was as follows: solid-liqu ilratio 1∶ 19,71% ethanol,refluxing and extracting for 128min,two times.The results indicated that D101 macroporous resins had better adsorption and desorption property for purification.The best condition was determined: Best mix sample than one;the biggest was a sample weight more than twenty;the elution solvent 3 BV of 70% ethanol,the flowrate at 2ml/min.Conclusion:This optimized process is economical,stable and reasonable.
